摘 要:目的观察艾拉莫德(Iguratimod,T-614)对干燥综合征模型小鼠(NOD小鼠)颌下腺组织炎性细胞浸润及血清细胞因子IL-17、IFN-γ水平的作用,探讨艾拉莫德对干燥综合征模型小鼠的影响。方法选取10周龄雌性NOD小鼠18只,随机分成3组:模型组、艾拉莫德1组、艾拉莫德2组;10周龄雌性C57BL/6小鼠6只,作为正常对照组。于实验第0天始,1组每只小鼠予10 mg/kg艾拉莫德灌胃治疗;2组予30 mg/kg艾拉莫德灌胃治疗;模型组和对照组每只小鼠予等体积0.5%羧甲基纤维素灌胃,每天灌胃1次,每2天测量1次小鼠进食量、饮水量、体重,直至鼠龄16周停止。16周龄时处死小鼠,眼球取血分离血清,ELISA测定血清中IL-17、IFN-γ水平,取颌下腺组织进行HE染色,光镜下评估各组小鼠外分泌腺组织炎性细胞浸润情况。结果与模型组相比2组IL-17在血清中水平显著减低(P <0.01);颌下腺组织炎性细胞浸润明显减轻;饮水量显著减少,进食量及体重明显增加(P <0.05);与1组比较2组颌下腺组织炎性细胞浸润有减轻。结论艾拉莫德能减轻外分泌腺淋巴细胞浸润程度,具有显著降低NOD小鼠血清IL-17水平,且随着剂量增加,效果更明显。艾拉莫德对模型小鼠外分泌腺组织具有保护作用以及改善外分泌腺功能。Objective To observe the effect of Iguratimod on the condition of lymphocytes infiltration in the salivary gland,cytokines interferon(IFN)-γ、interleukin(IL)-17 in serum of Sjogren′s Syndrome model mouse(NOD mouse),and investigate the possilbe mechanism of Iguratimod in the treatmeat of Sjogren′s Syndrome.Methods 18 female NOD mice which were ten-week-old,randomly divided into 3 groups:model group,10 mg/kg Iguratimod-treatment group(group1),30 mg/kg Iguratimod-treatment group(group2),6 female C57BL/6 mice which were also ten-week-old,considered normal control group.From the 0 day of the experiment,group 1 were treated with Iguratimod at a dose of 10 mg/kg for each mouse through oral gavage once a day;Group 2 were treated with Iguratimod at a dose of 30 mg/kg;Model group and normal control group were treated with the same volume of 0.5%carboxymethylcellulos.Food and water intake,bady weight were measured every 2 days.Untill the mice growth to 16 weeks.At 16-week-old,all the mice were sacrificed,and blood were collceted by extraction eyeball.ELISA was used to measure the level of IL-17、IFN-γin serum;HE staining was used to observe the lymphocytes infiltration condition in salivary gland under an inverted electron microscope.Result Compare to the model group,the level of IL-17 in serum are obviously lower in group 2(P<0.01),the conditions of cytokine infiltration in salivary gland obviously alleviate;Water intake is reduced,food intake and body weight are argumented(P<0.05).Compared to group 1,the conditions of cytokine infiltration in salivary gland alleviate in group 2.Conclusion Iguratimod can reduce the degree of lymphocytes infiltration in exocrine gland,and obviously reduc the level of IL-17 in serume of NOD mouse.So it has protective effect on the exocrine gland of mouse model and improve the excrine gland function.
