摘　　要：目的探究血管生成样蛋白2(ANGPTL2)水平变化对胃癌细胞增殖与侵袭的影响。方法选择2015年12月至2017年12月于我院住院的胃癌患者60例为研究对象,以其癌组织及癌旁组织作为此次研究的实验组与对照组。Western blot检测ANGPTL2表达水平,分析ANGPTL2含量与临床参数的关系,同时,分析GES-1、N87、SGC7901、BGC823、PAMC82胃细胞株中ANGPTL2的表达情况。过表达/沉默ANGPTL2后,MTT与Transwell试验检测ANGPTL2对细胞增殖率的影响。结果肿瘤恶性程度越高,ANGPTL2的表达水平也越高;Western Blot结果提示与GES-1相比,N87、SGC7901、BGC823、PAMC82胃癌细胞株中ANGPTL2的蛋白水平均呈较高水平(P <0.05);过表达ANGPTL2后,PAMC82胃癌细胞增殖能力及侵袭能力增强,而沉默ANGPTL2,其增值、侵袭能力减弱,差异有统计学意义(P <0.05)。结论 ANGPTL2在胃癌组织的表达水平较癌旁组织更高,且恶性程度越高ANGPTL2表达越高。其能加速胃癌细胞的增殖与侵袭,可作为临床治疗靶点之一。Objective To investigate the effect of ANGPTL2 levels on the proliferation and invasion in gastric cancer cells.Methods 60 patients with gastric cancer in our hospital from December 2015 to December 2017 were selected.The cancerous tissues and adjacent tissues were regard as experimental group and control group.Western blot was used to detect the expression level of ANGPTL2.The relationship between ANGPTL2 con-tent and clinical parameters was analyzed.At the same time,the expression of ANGPTL2 in GES-1,N87,SGC7901,BGC823 and PAMC82 gastric cell lines was analyzed.After overexpression/silence of ANGPTL2,MTT assay and Transwell assay were applied to examine the effect of ANGPTL2 on cell proliferation.Results The high-er the degree of tumor malignancy,the higher the expression level of ANGPTL2.Western Blot results suggested that the protein levels of ANGPTL2 in N87,SGC7901,BGC823 and PAMC82 gastric cancer cell lines were higher than those of GES-1(P<0.05).After overexpression of ANGPTL2,the proliferative capacity and invasive ability of PAMC82 gastric cancer cells were increased,but the value of ANGPTL2 was silenced and its invasion ability was weakened.The difference was statistically significant(P<0.05).Conclusion The expression level of ANGPTL2 in gastric cancer tissues is higher than that in adjacent tissues.The higher the degree of malignancy,the higher the expression of ANGPTL2,which could accelerate the proliferation and invasion of gastric cancer cells and could be used as one of the clinical therapeutic targets.
