GLUT1与MCT1在直肠癌组织中的表达及意义

摘　　要：目的探讨葡萄糖转运蛋白1(glucose transporter 1,GLUT1)和单羧酸转运蛋白1(mono-carboxylate transporters 1,MCT1)在人直肠癌组织中的表达及在肿瘤发生、发展中的意义。方法应用免疫组化法检测75例直肠癌及32例活检正常直肠组织标本(对照组)中GLUT1及MCT1的表达,分析GLUT1与MCT1在直肠癌组织中表达的关系,GLUT1、MCT1的表达与TNM分期及淋巴结转移的关系。结果 GLUT1表达阳性直肠癌组59例(78.67%),对照组1例(3.13%),差异有统计学意义(χ~2=51.97,P<0.05);GLUT1表达与直肠癌TNM分期及淋巴结转移有关(χ~2=9.252、6.460,均P<0.05)。MCT1表达阳性直肠癌组47例(62.67%),对照组2例(6.25%),差异有统计学意义(χ~2=28.76,P<0.05);MCT1表达与直肠癌TNM分期以及淋巴结转移有关(χ~2=9.744、9.316,均P<0.05)。随着直肠癌的进展,GLUT1和MCT1阳性表达率均呈上升趋势(Z=2.969、3.109,均P<0.01)。进一步分析发现,两者的表达水平在直肠癌组织中呈线性相关(r=0.639,P<0.05)。结论 GLUT1与MCT1的表达在直肠癌发生、发展过程中可能存在一定的协同作用。Objective To investigate the expression and clinical significance of glucose transporter 1(GLUT1)and Mono-carboxylate transporters 1(MCT1)in patients with rectal cancer.Methods Immunohistochemical method was used to detect the expression of GLut1 and MCT1 in rectal tissues of 75 rectal cancer patients and 32 controls.The relationship between expression of GLUT1 and MCT1,the relationship between expression of GLUT1 and MCT1 with TNM staging and lymph node metastasis in rectal cancer were analyzed.Results The expression rate of GLUT1 in cancer patients was significantly higher than that in controls(78.67%vs.3.13%,χ^2=51.97,P<0.05).The positive expression of GLUT1 was correlated with TNM staging and lymph node metastasis(χ^2=9.252,6.460,P<0.05).The positive expression rate of MCT1 in cancer patients was significantly higher than that in controls(62.67%vs.6.25%,χ^2=28.76,P<0.05).The expression of MCT1 was correlated with TNM staging and lymph node metastasis(χ^2=9.744,9.316,P<0.05).With the development of rectal cancer,the positive expression rates of GLUT1 and MCT1 showed an upward trend.Further analysis found that the expression levels of both proteins were linearly correlated in rectal cancer tissue(r=0.639,P<0.05).Conclusion The expression of GLUT1 and MCT1 may have a synergistic effect in the occurence and develop of rectal cancer.
