摘  要:[目的]提高马铃薯茎尖组织培养的成苗率和脱毒率,对马铃薯组织培养技术进行优化。[方法]首先从马铃薯幼苗上取生长健壮的芽,经消毒后取1.5 mm左右茎尖,接种于分化培养基上;随后分析不同茎尖大小、不同浓度和配比的激素对马铃薯茎尖的诱导效果。[结果]茎尖越小,脱毒率越高,但成活率和成苗率越低,以1.5 mm左右茎尖脱毒效果最好,成苗率也高;低浓度的GA3可促进愈伤组织的形成,过高的浓度会抑制愈伤组织的形成。[结论]在MS+0.5 mg/L 6-BA+0.2 mg/L GA3+0.1 mg/L NAA培养基上马铃薯茎尖组织诱导效果较好。[Objective]The aim was to improve the seedling rate and virus-free rate of stem tip tissue culture of potato and optimize the potato tissue culture technology.[Method]First,the healthy growing buds were taken from the potato seedlings,and about 1.5 mm stem tip was taken after disinfection and inoculated on the differentiation medium.Then,the induction effects of different stem tip sizes,different concentrations and ratios of hormones on potato stem tips were analyzed.[Result]The results showed that the smaller the stem tip,the higher the virus-free rate,but the lower the survival rate and the seedling rate.The virus-free effect and the seedling rate of the stem tip with length 1.5 mm were the best.The low concentration of GA3 could promote the formation of callus,and the high concentration could inhibit the formation of callus.[Conclusion]The MS+0.5 mg/L 6-BA+0.2 mg/L GA3+0.1 mg/L NAA was the best medium for inducing potato shoot tip tissue.
