摘　　要：目的探讨微小RNA-378(miR-378)在乳腺癌组织中的表达及临床意义。方法收集2012年1月至2014年12月女性乳腺癌患者手术切除的105对癌组织和癌旁组织,采用实时荧光定量PCR(QPCR)检测上述组织中miR-378的表达并比较miR-378在癌组织和癌旁组织中的分布差异,进一步分析miR-378表达与乳腺癌临床病理特征的关系,根据随访资料分析不同miR-378水平患者的预后情况。结果 miR-378在乳腺癌组织中的表达水平为2. 745±0. 347,高于癌旁组织的1. 085±0. 126(P<0. 01)。miR-378水平与年龄、淋巴结转移、绝经、组织学分级、ER和PR表达均无关,而与肿瘤大小、TNM分期、HER-2表达、Ki-67表达和Nottingham预后指数有关(P<0. 05)。单因素分析发现,乳腺癌患者的总生存期(OS)与年龄、肿瘤大小、绝经、ER和PR表达均无关,而与淋巴结转移、TNM分期、组织学分级、HER-2表达、Ki-67表达、Nottingham预后指数和miR-378水平有关(P<0. 05); miR-378低表达者的中位OS为>60. 0个月,高于高表达者的41. 0个月,差异有统计学意义(P<0. 05)。结论 miR-378在乳腺癌中高表达且与肿瘤的发生、发展和预后有一定关系,在乳腺癌筛查及预后预测上有一定的潜在价值。Objective To investigate the expression of microRNA-378(miR-378)in breast cancer tissues and its clinical significance.Methods From January2012to December2014,105pairs of cancerous and paracancerous tissues surgically removed from105patients with breast cancer were collected.Real-time fluorescence quantitative PCR(QPCR)was used to detect the expression of miR-378in the above tissues and the distribution of miR-378in cancerous tissues and paracancerous tissues were compared.The relationship between the expression of miR-378and clinicopathological parameters of breast cancer was further analyzed.According to the follow-up data,the prognosis of different miR-378levels was analyzed.Results Compared with paracancerous tissues,miR-378expression was upregulated in breast cancer tissue(2.745±0.347vs.1.085±0.126,P<0.01).MiR-378level was not related with age,lymph node metastasis,menopause,histological grade,ER and PR expression,but with tumor size,TNM stage,HER-2expression,Ki-67expression and Nottingham prognosis index(P<0.05).Univariate analysis showed that overall survival(OS)was not associated with age,tumor size,menopause,ER and PR expression,but with lymph node metastasis,TNM stage,histological grade,HER-2expression,Ki-67expression,Nottingham prognostic index and miR-378expression(P<0.05).The median OS of patients with low expression of miR-378was more than60.0months,better than41months of patients with high expression of miR-378(P<0.05).Conclusion MiR-378is highly expressed in breast cancer and related to tumor progress and prognosis in breast cancer.These findings indicate that miR-378may be regarded as a potential biomarker for the detection of breast cancer.
