摘 要:目的探讨多发性骨髓瘤(MM)患者骨髓组织中微小RNA-181a(miR-181a)和微小RNA-373(miR-373)的表达及临床意义。方法收集2013年1月至2018年3月收治的MM患者64例(MM组),并选取同时期进行骨髓穿刺并最后明确骨髓功能无异常的健康者64例(正常对照组)。采用实时荧光定量PCR(QPCR)检测两组骨髓组织中miR-181a、miR-373的表达水平,并分析两者与MM临床病理特征的关系;采用Pearson法分析miR-181a和miR-373表达的相关性。结果 MM组骨髓组织中miR-181a的表达水平为112. 68±16. 53,高于正常对照组的75. 94±10. 02,差异有统计学意义(P<0. 01); miR-373的表达水平为0. 36±0. 06,低于正常对照组的1. 12±0. 10,差异有统计学意义(P<0. 01)。MM患者骨髓组织中miR-181a和miR-373的表达呈负相关(r=-0. 928,P<0. 001)。miR-181a和miR-373表达水平与MM患者的年龄、临床分期、分化程度及淋巴结转移有关(P<0. 001),与性别及一般分型无关(P>0. 05)。结论 miR-181a、miR-373在MM骨髓组织中分别为高表达和低表达,两者表达存在负相关,并与临床分期、淋巴结转移和分化程度有关,可作为MM潜在的分子筛查标志物。Objective To investigate the expression and clinical significance of microRNA-181a(miR-181a)and microRNA-373(miR-373)in bone marrow of patients with multiple myeloma(MM).Methods Sixty-four MM patients(MM group)from January2013to March2018were selected.Sixty-four healthy subjects(normal control group)were selected who underwent bone marrow biopsy at the same time and were finally identified as normality in bone marrow function.The expression levels of miR-181a and miR-373in the two groups of bone marrow tissues were detected by real-time fluorescence quantitative PCR(QPCR).The relationship between expression of miR-181a and miR-373and clinicopathological parameters of MM was analyzed.Pearson correlation analysis was used to analyze the correlation between miR-181a and miR-373expression.Results The expression of miR-181a in bone marrow tissues of MM group was112.68±16.53,higher than75.94±10.02of normal control group(P<0.01).The expression of miR-373in bone marrow tissue of MM group was0.36±0.06,lower than1.12±0.10of normal control group(P<0.01).There was a negative correlation between the expression of miR-181a and miR-373in bone marrow tissue of MM patients(r=-0.928,P<0.001).The expression levels of miR-181a and miR-373were correlated with age,clinical stage,differentiation and lymph node metastasis(P<0.001),but not with gender and general type(P>0.05).Conclusion MiR-181a is highly expressed in MM bone marrow tissues,while miR-373is abnormally low.There is a negative correlation between their expression and they are correlated with clinical stage,lymph node metastasis and differentiation,which can be used as potential molecular screening markers for MM.
