不同剂量对药酸枣仁-合欢花对抑郁模型大鼠体重及行为学指标的影响

摘　　要：目的观察不同剂量对药酸枣仁-合欢花对抑郁模型大鼠体重及行为学指标的影响。方法将48只SD大鼠随机分为正常组、模型组、盐酸文拉法辛组及不同剂量(4 g/kg、8 g/kg、16 g/kg)对药酸枣仁-合欢花组,每组各8只。除正常组外,其他5组采用孤养加慢性不可预见性应激的方法建立抑郁模型。造模的同时正常组及模型组给予蒸馏水灌胃,其他组给予相应的药物灌胃。于建模前1 d和给药后1、2、3周,比较6组大鼠的体重、糖水消耗率、游泳不动时间。结果给药后2、3周,各给药组大鼠体重及糖水消耗率均高于模型组,而游泳不动时间短于模型组(均P <0. 05)。在给药后3周,8 g/kg对药酸枣仁-合欢花组大鼠体重高于其他给药组,而游泳不动时间短于其他剂量对药酸枣仁-合欢花组,糖水消耗率高于其他剂量对药酸枣仁-合欢花组(均P <0. 05)。结论对药酸枣仁-合欢花能增加抑郁模型大鼠的体重,改善行为学指标,且8 g/kg对药酸枣仁-合欢花的效果更佳。Objective To observe the effects of different doses of herbal pair of Semen Ziziphi Spinosae(SZS)and Albizia Julibrissin Flower(AJF)on body weight and ethological indices in a rat model of depression.Methods Forty-eight SD rats were randomly divided into normal group(n=8),model group,venlafaxine hydrochloride group,and SZS-AJF groups in which different doses(4g/kg,8g/kg and16g/kg)of herbal pair of SZS and AJF were used,respectively,with8rats in each group.Except the normal group,the rats in the other5groups were raised in isolation to develop the depression model using the method of chronic unpredictable stress.The normal group and model group were given distilled water by gavage when modeling,while other groups were given corresponding drugs by gavage.One day before modeling and1,2and3weeks after medication,body weight,sugar water consumption rate,and immobility time in forced swimming test were compared among the six groups.Results Two and3weeks after medication,the rats in each drug group had higher body weight and sugar water consumption rate as well as less immobility time in forced swimming test compared with the model group(all P<0.05).Three weeks after medication,the rats in the8g/kg SZS-AJF group had higher body weight compared with other drug groups,and got less immobility time in forced swimming test and higher sugar water consumption rate compared with the SZS-AJF groups of other doses(all P<0.05).Conclusion The herbal pair of SZS and AJF can increase body weight and improve the ethological indices in a rat model of depression,and the herbal pair of SZS and AJF(8g/kg)achieves better efficacy.
