摘 要:目的观察雷公藤内脂醇作用于去势抵抗前列腺癌PC3细胞后对其生长增殖的影响。方法 PC3细胞培养后,分别用浓度为5、10、20、40、80、160 nmol/L的雷公藤内脂醇培养液作用于PC3细胞,用MTT法检测细胞生长增殖的变化;用流式细胞仪检测细胞的凋亡的变化;用ELISA法检测VEGF的含量。结果用含有雷公藤内脂醇的培养液培养PC3细胞后,细胞的贴壁功能慢慢减退,PC3细胞出现细胞膜皱缩、染色质浓缩、核膜核仁消失、核固缩、出现空泡等现象。随着雷公藤内酯醇剂量的增加和培养时间的延长,对PC3细胞增殖的抑制作用越明显(P <0. 05),雷公藤内脂醇对PC3细胞的凋亡呈现出时间与浓度的依赖性。含量为10、20、40 nmol/L浓度的雷公藤内脂醇细胞培养上清中VEGF含量分别为(415. 24±22. 73)%、(323. 57±28. 26)%、(251. 67±32. 61)%,与对照组(465. 48±30. 19)%相比,差异有统计学意义(P <0. 05)。结论雷公藤内脂醇作用于PC3细胞后可以下调肿瘤细胞VEGF的含量,抑制肿瘤血管的新生,从而诱导细胞凋亡,抑制细胞生长增殖。Objective To observe the effect of triptolide on the growth and proliferation of prostate cancer PC3 cells after ovariectomy.Methods PC3 cells were cultured in PC3 cells with 5,10,20,40,80 and 160 nmol/L triptolide,respectively.The growth and proliferation of PC3 cells was detected by MTT assay.The changes of apoptosis were detected by ELISA.The content of VEGF was detected by ELISA.Results After the PC3 cells were cultured with the culture medium containing triptolide,the adherent function of the cells gradually decreased.The cell membrane shrinkage,chromatin condensation,nuclear nucleolus disappearance,nuclear condensation,vacuolization,etc.phenomenon.With the increase of triptolide and the prolongation of culture time,the inhibitory effect on the proliferation of PC3 cells was more significant(P<0.05).The apoptosis of PC3 cells was dependent on the time and concentration.(415.24±22.73)%,(323.57±28.26)%and(251.67±32.61)%,respectively,compared with the control group(465.48±30.19)%,and the content of VEGF in the supernatant of triptolide was 10,20,40 nmol/L,the difference was statistically significant(P<0.05).Conclusion The effect of triptolide on PC3 cells can down-regulate the VEGF content of tumor cells and inhibit the neovascularization of tumor cells,thus induces apoptosis and inhibit cell growth and proliferation.
