痉挛型脑瘫双生子差异表达基因筛选及功能分析

摘  要:目的利用基因表达谱芯片筛选出痉挛型脑瘫双生子患儿差异表达基因。方法选择痉挛型脑瘫—正常双生子两对作为研究对象,其中两例痉挛型脑瘫作为对照组,两正常儿童做为空白组。分别抽取各组受检者外周静脉血5 m L,用TRIzol法提取总RNA后采用Human Genome Array表达谱芯片筛选差异表达基因,利用DAVID生物软件对差异表达基因进行GO富集和KEGG通路分析。结果筛选出13个特异性差异表达基因,这些基因主要参与了免疫、细胞分化、肌肉张力及蛋白翻译合成等调节。结论痉挛型脑瘫双生子存在差异表达的基因,这些基因可能是痉挛型脑瘫表型发生发展的关键病机之一。Objective To screen differentially expressed genes in children with spastic cerebral palsy twin by using gene expression microarray.Methods Two pairs of spastic cerebral palsy-normal twins were selected as study subjects,including two spastic cerebral palsy as control group and two normal children as blank group.The peripheral venous blood of each group was drawn5mL,the total RNA was extracted by TRIzol method,and the differentially expressed genes were screened by Human Genome Array.The GA gene expression and KEGG pathway were analyzed by DAVID software.Results Thirteen specific differentially expressed genes were selected and these genes were mainly involved in the regulation of immunity,cell differentiation,muscle tone and protein translation and synthesis.Conclusion There are differentially expressed genes in twins of spastic cerebral palsy.These genes may be one of the key pathogenesis of spastic cerebral palsy phenotype.
