摘　　要：【目的】研究胰腺癌患者脱氧胞苷激酶(deoxyeytidine kinase,dCK)A9846G位点单核苷酸多态性(single nucleotide polymorphism,SNP)分布情况,并探索其与吉西他滨化疗预后的关系。【方法】应用DNA直接测序法检测75例根治性切除的胰腺癌患者组织中dCK基因A9846G位点SNP序列,分析dCK基因SNP对胰腺癌患者吉西他滨化疗生存期的影响。【结果】75例胰腺癌患者dCK基因A9846G中野生型AA有14例、杂合型AG有38例,突变型GG有23例。dCK基因A9846G位点基因型与胰腺癌患者临床病理因素无相关性(P>0.05)。dCK基因A9846G位点AA基因型携带者较AG或GG基因型携带者对吉西他滨的化疗反应更敏感,患者总生存率和无病生存率更高(P<0.05)。【结论】可切除性胰腺癌患者dCK基因A9846G SNP与胰腺癌患者吉西他滨化疗预后有明显相关性,可能成为有效预判胰腺癌患者吉西他滨化疗预后的敏感指标。【Objective】To investigate the distribution of single nucleotide polymorphism(SNP)in dCK gene A9846G in patients with pancreatic cancer and to explore its relevance with the prognosis of gemcitabine chemotherapy.【Methods】The SNP sequence of dCK gene was detected by direct DNA sequencing in 75 patients with pancreatic cancer undergoing radical resection.Furtheremore,the relevance of dCK gene A9846G SNP and the survival time of pancreatic cancer patients with gemcitabine chemotherapy was analyzed.【Results】In patients with pancreatic cancer,the wild-type AA and heterozygous AG and mutant GG were 14,38 and 23 in dCK gene A9846G,respectively.There was no correlation between A9846G genotype of dCK gene and clinicopathological factors in patients with pancreatic cancer(P>0.05).The carriers of genotype AA were more sensitive to gemcitabine chemotherapy than those patients with AG or GG genotype,and the overall survival rate and disease-free survival rate were higher(P<0.05).【Conclusion】The dCK gene A9846G SNP is significantly correlated with the prognosis of gemcitabine chemotherapy in patients with pancreatic cancer which may be a sensitive indicator for predicting the prognosis of gemcitabine chemotherapy in patients with pancreatic cancer.
