宫颈癌组织TWIST与E-Cadherin的表达水平及相关性分析

摘　　要：目的探究宫颈癌组织TWIST与E-Cadherin的表达水平及其相关性。方法选取2014年7月—2017年6月行全子宫切除术联合盆腔淋巴结清扫术治疗患者的完整宫颈癌标本74例作为宫颈癌组,同期行Leep手术切除或阴道下活检的完整宫颈上皮内瘤变(CIN)标本35例作为CIN组,以及同期因子宫肌瘤行子宫全切术治疗的正常宫颈组织22例作为正常宫颈组。检测TWIST与E-Cadherin蛋白的表达水平,分析其与宫颈癌临床病理特征的关系,并分析TWIST与E-Cadherin表达水平的相关性。结果宫颈癌组的TWIST阳性表达率高于正常宫颈组和CIN组,ECadherin阳性表达率低于正常宫颈组和CIN组(P <0. 05)。与间质浸润深度<1/2患者比较,间质浸润深度≥1/2患者宫颈癌组织中TWIST的阳性表达率升高,E-Cadherin阳性表达率降低(P <0. 05)。与无淋巴结转移患者比较,有淋巴结转移患者宫颈癌组织中TWIST的阳性表达率升高,E-Cadherin阳性表达率降低(P <0. 05)。TWIST与E-Cadherin在宫颈癌组织中表达水平呈负相关(r=-0. 511,P <0. 001)。结论 TWIST表达升高与E-Cadherin表达降低可能与宫颈癌的发生发展及浸润转移密切相关,二者可能具有拮抗作用。Objective To investigate expressions of TWIST and E-Cadherin and their correlation in cervical cancer tissues.Methods A total of 74 completely cervical cancer cases from patients undergoing complete hysterectomy combined with pelvic lymphadenectomy during July 2014 and June 2017 were selected as cervical cancer group,while other 35 completely cervical intraepithelial neoplasia(CIN)specimens from patients undergoing Leep resection or subvaginal biopsy simultaneously were selected as CIN group,and other 22 cases of normal cervical tissues from patients undergoing total hysterectomy for uterine fibroids at the same period were selected as normal cervical group.Expression levels of TWIST and E-Cadherin were detected,and correlations between the expressions with clinical pathological features were analyzed,and correlation between TWIST with E-Cadherin was analyzed.Results In cervical cancer group,TWIST positive expression rate was significantly higher,while E-Cadherin positive expression rate was significantly lower than those in normal cervical cancer and CIN groups(P<0.05).Compared with those in cervical cancer patients with interstitial infiltration depth less than 1/2,TWIST positive expression rate was significantly higher,while E-Cadherin positive expression rate was significantly lower in cervical cancer patients with interstitial infiltration depth equal or larger than 1/2(P<0.05).Compared with those in cervical cancer patients without lymph node metastasis,TWIST positive expression rate was significantly increased,while E-Cadherin positive expression rate was significantly decreased in cervical cancer patients with lymph node metastasis(P<0.05).TWIST and E-Cadherin expressions were negatively correlated in patients with cervical cancer(r=-0.511,P<0.001).Conclusion Increased expression of TWIST and decreased expression of E-Cadherin in patients with cervical cancer may be closely related to pathogenesy,development,invasion and metastasis of cervical cancer,and antagonistic effects may be existed in them.
