摘 要:目的探讨PI3K/Akt信号通路与放射性肝损伤的关系。方法选取大白兔20只,随机分为观察组(n=10)和对照组(n=10),两组大白兔再随机分为照射前组和照射后7 d组,各5只。观察组给予右半肝伽玛刀单次照射,对照组给予假照射,检测两组大白兔肝功能,采用HE染色和Masson染色观察肝脏组织结构,免疫组化染色检测TGF-β1、α-SMA和p-Akt表达。结果观察组照射后7 d谷丙转氨酶(AST)、谷草转氨酶(ALT)、碱性磷酸酶(ALP)和总胆红素(TB)分别为(164.51±25.64)、(76.68±12.10)、(301.15±40.41)和(5.68±0.41)μmol/L,明显高于照射前及对照组(P<0.05);照射后7 d,观察组大白兔肝细胞排列紊乱,部分细胞胞质疏松,脂肪变性,呈空泡状,肝小叶结构紊乱,肝小叶内逐渐呈纤维间隔;观察组照射后7 d TGF-β1、α-SMA和p-Akt表达吸光值分别为(0.087±0.012)、(0.073±0.010)和(0.075±0.014),明显高于照射前及对照组(P<0.05)。结论 PI3K/Akt信号通路可能参与了放射性肝损伤过程,值得进一步研究。Objective To investigate the relationships between PI3K/Akt signaling pathway and radiation-induced liver injury.Methods Twenty rabbits were randomly divided into the observation group( n = 10) and control group( n = 10). The two groups of rabbits were again randomly divided into the pre irradiation group and the 7 d after irradiation group,with 5 rabbits in each. The observation group was given the right liver gamma knife single exposure,while the control group was given false irradiation. Hepatic function was detected in the two groups. The histological structure of the liver was observed by HE staining and Mason staining,while the expressions of TGF-β1,α-SMA and p-Akt were detected by immunohistochemical staining.Results The levels of alanine aminotransferase( AST),glutamic oxaloacetic aminotransferase( ALT),alkaline phosphatase( ALP) and total bilirubin( TB) in the observation group were( 164.51±25.64) mmol/L,( 76.68±12.10) mmol/L,( 301.1± 40.41) mmol/L and( 5.68±0.41) mmol/L respectively,significantly higher than those before irradiation and those of the control group( P< 0.05). On the 7 th day after irradiation,the hepatocytes in the observation group were disordered,some of the cells were loose and steatosis and vacuole-like,the structure of hepatic lobules was disordered,and the hepatic lobules gradually showed fibrous septa. The expressions of TGF-β1,α-SMA and p-Akt in the observation group at 7 days after irradiation were( 0.087±0.012),( 0.073± 0.010)and( 0.075±0.014) respectively,which were significantly higher than those before irradiation and in the control group( P<0.05).Conclusion PI3 K/Akt signaling pathway may be involved in the process of radiation-induced liver injury,which is worthy of further study.
