摘 要:目的:研究豌豆根瘤菌RL3841的谷胱甘肽还原酶编码基因gshR的功能.方法:采用基因同源重组构建了豌豆根瘤菌gshR基因突变株RLgshR,探讨了基因突变对根瘤菌抗氧化和共生固氮的影响,利用实时荧光定量RTPCR检测gshR基因的mRNA表达水平.结果:gshR基因缺失不影响菌株在AMS基础培养基中的生长能力,但突变株对氢过氧化枯(CuOOH)和较高浓度的H_2O_2氧化物敏感.结论:gshR基因突变株RLgshR形成正常固氮根瘤,gshR基因的表达不受H_2O_2和共生环境诱导.Objective:To study the function of glutathione reductase gene gshR in rhizobium leguminosarum.Method:gshR mutant RLgshR was constructed through homologous recombination,and the effect of gene mutation on its symbiosis nitrogen fixation and antioxidative function were explored.Quantitative real-time RT-PCR was performed to detect mRNA expression of gshR gene.Results:The growth ability of RLgshR in AMS basal medium was not affected by the lack of gshR gene;however,the mutant strain was sensitive to CuOOH and high concentrations of H 2 O 2.Conclusion:The gshR mutant RLgshR formed nodules with normal nitrogen-fixing activity and gshR gene expression can not be induced under H 2 O 2 stress and symbiotic condition.
