甘肃棘豆黄酮抗氧化活性及免疫活性的研究

摘　　要：为研究甘肃棘豆黄酮的抗氧化及免疫活性,试验通过1,1-二苯基-2-三硝基苯肼(DPPH)自由基、超氧阴离子自由基和羟基自由基的清除率测定其抗氧化活性,通过测定不同浓度甘肃棘豆黄酮对小鼠脾脏淋巴细胞增殖能力、白细胞介素2(IL-2)和免疫球蛋白G(IgG)抗体形成的影响研究其免疫活性。结果显示,甘肃棘豆黄酮对DPPH自由基清除率最高为32.23%,略高于维生素E(28.55%),低于维生素C(97.05%);对超氧阴离子自由基清除率最高为80.03%,高于维生素E(69.70%),低于维生素C(98.69%);对羟基自由基清除率最高为30.39%,低于维生素C(98.98%)、略低于维生素E(32.57%)。在小鼠脾脏淋巴细胞增殖试验中,随着甘肃棘豆黄酮溶液浓度的提高,脾脏淋巴细胞相对增殖率也提高,当其浓度为200μg/mL时,小鼠脾脏淋巴细胞的增殖率为23.40%;200μg/mL甘肃棘豆黄酮组IL-2水平极显著高于对照组(P<0.01),50和100μg/mL甘肃棘豆黄酮组与对照组无显著差异(P>0.05);各甘肃棘豆黄酮组IgG抗体水平均极显著高于对照组(P<0.01),其中,50μg/mL甘肃棘豆黄酮组极显著高于100和200μg/mL甘肃棘豆黄酮组(P<0.01),100μg/mL甘肃棘豆黄酮组极显著高于200μg/mL甘肃棘豆黄酮组(P<0.01)。综上所述,甘肃棘豆黄酮具有较好的抗氧化活性及免疫活性。To investigate the antioxidation and immune activity of Oxytropis kansuensis flavonoids,the antioxidant activities of Oxytropis kansuensis flavonoids were determined by DPPH,superoxide anion and hydroxyl free radical scavenging activities,and the immunomodulatory of Oxytropis kansuensis flavonoids were studied by measuring the effects of different concentrations of Oxytropis kansuensis flavonoids on the proliferation of mouse spleen lymphocytes,the concentrations of IL-2and IgG.The results showed that the highest scavenging rate of DPPH free radicals of Oxytropis kansuensis flavonoids was32.23%which was slightly higher than vitamin E(28.55%)and lower than vitamin C(97.05%).The highest scavenging rate of superoxide anion free radicals of Oxytropis kansuensis flavonoids was80.03%which was higher than vitamin E(69.70%)and lower than vitamin C(98.69%).The highest clearance rate of hydroxyl free radicals of Oxytropis kansuensis flavonoids was30.39%which was lower than that of vitamin C(98.98%)and slightly lower than that of vitamin E(32.57%).In the spleen lymphocyte proliferation assay on mice,the relative proliferation rate of spleen lymphocytes increased with the increase concentration of Oxytropis kansuensis flavonoids.When the concentration of Oxytropis kansuensis flavonoids was200μg/mL,the spleen lymphocyte proliferation rate was23.40%.The IL-2concentration in200μg/mL Oxytropis kansuensis flavonoids group was extremely significantly higher than that of control group(P<0.01),and there was no significant difference between50,100μg/mL Oxytropis kansuensis flavonoids groups and control group(P>0.05);The IgG antibody concentration of Oxytropis kansuensis flavonoids groups were extremely significantly higher than that of control group(P<0.01),50μg/mL Oxytropis kansuensis flavonoids group was extremely significantly higher than that of100and200μg/mL Oxytropis kansuensis flavonoids groups(P<0.01),and100μg/mL Oxytropis kansuensis flavonoids group was extremely significantly higher than200μg/mL Oxytropis kansuensis
