摘 要:目前乳腺癌是女性最常发病的恶性肿瘤之一,其中三阴性乳腺癌(TNBC)是恶性程度较高的亚型。TNBC因雌激素受体、孕激素受体、人类表皮生长因子受体2表达均为阴性,缺乏内分泌治疗及靶向治疗的常规手段,故其治疗更是成为当今医学领域的热点、难点。乳腺癌易感基因1(BRCA-1)是最早发现也是最主要的乳腺癌抑癌基因,近年来研究发现BRCA-1突变与TNBC的发生、发展密切相关。此外,微RNA在转录后水平对基因表达进行负性调控,发现在癌症中充当致癌基因和肿瘤抑制基因,并在TNBC中表达异常,通过多种分子机制促进TNBC细胞的增殖、侵袭及转移。At present,breast cancer is one of the most common malignant tumors in women today.Three-negative breast cancer(TNBC)is a subtype with a higher degree of malignancy.TNBC is negative of estrogen receptor,progesterone receptor and human epithelial growth factor receptor-2expression,and lacks conventional methods of endocrine therapy and targeted therapy.Therefore,its treatment has become a hot and difficult issue in the medical field.Breast cancer susceptibility gene1(BRCA-1)is the earliest discovered and first major breast cancer suppressor gene.In recent years,it′s found that BRCA-1mutation is closely related to the occurrence and development of TNBC.In addition,microRNA negatively regulate gene expression on post-transcriptional level,acting as both oncogenes and tumor suppressor genes in cancer,with abnormal expression in TNBC,promoting proliferation,invasion and metastasis of TNBC cells through various molecular mechanisms.
