摘  要:目的探究miR-181b-3p对胆囊癌细胞侵袭转移能力的影响并初步探讨其机制。方法采用real time PCR检测胆囊癌组织及正常组织中miR-181b-3p的含量;通过Transwell实验检测miR-181b-3p对胆囊癌细胞侵袭转移能力的影响;Western blot检测miR-181b-3p对E-钙黏蛋白、波形蛋白表达的影响,并探究miR-181b-3p与E-钙黏蛋白表达的相关关系。结果 PCR显示胆囊癌组织中miR-181b-3p表达量(9.22±5.74)明显高于正常组织(2.54±1.27)(t=3.213,P=0.006 3)。Transwell实验表明miR-181b-3p过表达组侵袭转移的细胞数量为(811.40±179.53)个,明显高于对照组的(231.00±61.22)个(LSD-t=8.357,P=0.000 9);而miR-181b-3p下调组侵袭转移的细胞数量为(28.80±13.81)个,明显低于其对照组的(231.00±61.22)个(LSD-t=2.912,P=0.001 2)。Western blot实验表明上调miR-181b-3p使胆囊癌细胞E-钙黏蛋白表达减少,波形蛋白表达增加,下调则得到相反结果;通过统计学分析表明,胆囊癌组织中E-钙黏蛋白表达量与miR-181b-3p含量存在负相关关系。结论 miR-181b-3p通过促进胆囊癌细胞的上皮-间充质转化,增强了其侵袭转移能力,提示miR-181b-3p在胆囊癌侵袭转移过程中起着重要作用,有望成为胆囊癌治疗的潜在靶点。Objective To investigate the effects of miR-181b-3p on cell invasion and metastasis in gallbladder cancer cells and its mechanisms.Methods PCR was used to detect the level of miR-181b-3p in gallbladder cancer tissue and peritumoral tissues;Transwell assay to detect the effects of miR-181b-3p on the invasion and metastasis ability of gallbladder cancer cells and Western blot to detect the influence of miR-181b-3p on the expression of E-cadherin and vimentin.The correlation of miR-181b-3p with E-cadherin expression level was analyzed.Results PCR showed that miR-181b-3p level was higher in gallbladder cancer tissues(9.22±5.74)than that in peritumoral tissues(2.54±1.27)(t=3.213,P<0.05).Transwell assay showed that the number of transmembrane cells of up-regulated miR-181b-3p group was(811.40±179.53),significantly larger than that in the control group(231.00±61.22)(LSD-t=8.357,P<0.001)and that of down-regulated miR-181b-3p group was(28.80±13.81),significantly smaller than that of the control group(231.00±61.22)(LSD-t=2.912,P<0.05).Western blot showed up-regulation of miR-181b-3p decreased the expression of E-cadherin and increased the expression of vimentin in gallbladder cancer cells while down-regulation group showed the opposite results.Statistical analysis showed that there was a negative correlation between miR-181b-3p and E-cadherin in gallbladder cancer tissues.Conclusions miR-181b-3p increases the invasion and metastasis ability of gallblad-der cancer cells by promoting its epithelial-mesenchymal transition(EMT).miR-181b-3p plays an important role in the invasion and metastasis process of gallbladder cancer cells and would be a potential target for the treatment of gallbladder cancer.
