摘  要:目的观察多发性骨髓瘤患者血清蛋白质谱的变化,筛选出特异的蛋白标志物,并分析这些差异蛋白在多发性骨髓瘤患者中的意义。方法利用CM10蛋白芯片和SELDI-TOF-MS技术对32例初诊为多发性骨髓瘤的患者和35例健康人的血清蛋白进行分析。获得的蛋白质谱图采用Ciphergen公司的Biomarker Wizard软件分析。结果通过对多发性骨髓瘤患者血清与健康人血清蛋白质谱图分析发现有28个蛋白峰表达量有明显差异(P<0.05),与健康对照组相比,10个蛋白峰表达上调,18个蛋白峰表达下调,质荷比为33425.65、6582.57、5632.14、4874.12、7845.16的蛋白更具意义。结论结果表明通过多发性骨髓瘤患者与健康对照血清蛋白质谱的比较,有助于筛选得到多发性骨髓瘤的特异性标志物。Objective To observe the changes of serum protein mass spectrometry in patients with multiple myeloma,to screen out specific protein markers and to analyze the significance of these differential proteins in patients with multiple myeloma.Methods The serum proteins of 32 newly diagnosed multiple myeloma patients and 35 healthy people were analyzed by CM10 protein chip and SELDI-TOF-MS technique.The protein mass spectrum obtained was analyzed by Ciphergen BiomarkerW Izard software.Results The expression of 28 protein peaks in the serum and healthy people of multiple myeloma patients was found to be significantly different(P<0.05).Compared with the healthy control group,the expression of 10 protein peaks was up,18 protein peaks were down regulated,and the protein of 33425.65,6582.57,5632.14,4874.12 and 7845.16 was more significant.Conclusion The results showed that the comparison of serum protein mass spectrometry between patients with multiple myeloma and healthy control was helpful to screen the specific markers of multiple myeloma.
