摘 要:目的建立文冠果果壳醇提物总多酚含量(TPC)测定方法,并评价文冠果果壳醇提物不同部位总多酚含量和体外抗氧化活性。方法采用分光光度法测定文冠果果壳总多酚的含量;采用ABTS和DPPH自由基评价文冠果果壳多酚的抗氧化活性。结果文冠果果壳总多酚含量为10.22 mg/g。D101大孔树脂纯化后,多酚主要集中在30%乙醇洗脱部位(4.92 mg/g)。该部位ABTS和DPPH自由基的IC_(50)值分别为(4.83±0.33)、(9.33±0.23)μg/mL;阳性对照芦丁对ABTS和DPPH自由基的IC_(50)值为(12.87±0.21)、(10.29±0.17)μg/mL。结论文冠果果壳醇提物大孔树脂30%乙醇洗脱部位的总多酚含量最高,抗氧化活性最强,值得进一步开发利用。Objective To establish the determination method of total polyphenol content(TPC)of the ethanol extract from X.sorbifolia husks,and evaluate the TPC and antioxidant activity in vitro of different ethanol fractions from X.sorbifolia husks.Methods The TPC of X.sorbifolia husks was analyzed by spectrophotometry.The antioxidant activity of different ethanol fractions from X.sorbifolia husks was evaluated by ABTS and DPPH methods.Results The TPC of X.sorbifolia husks was10.22mg/g.After purified by D101macroporous resin,the polyphenols from X.sorbifolia husks were mainly concentrated in the30%ethanol fraction,the TPC of which was4.92mg/g.This faction exhibited strong antioxidant activity,the IC50values against ABTS and DPPH free radicals of which were(4.83±0.33),(9.33±0.23)μg/mL respectively.While the IC50values of the positive control rutin against ABTS and DPPH free radicals were(12.87±0.21),(10.29±0.17)μg/mL respectively.Conclusion The30%ethanol fraction purified by D101macroporous resin has the highest TPC and the strongest antioxidant activity,which is worthy of further development and utilization.
