摘  要:以福鼎大白茶(Camellia sinensis cv.Fuding-dabaicha)花粉为研究对象,考虑不同浓度的蔗糖、硼酸、硝酸钙,采用正交试验设计优化茶树花粉离体萌发培养基;通过离体萌发法、TTC染色法的对比,探讨茶树(C.sinensis)花粉活力的快速测定方法。结果表明,硼酸是影响茶树花粉离体萌发效果的主要因素,而不同浓度蔗糖的影响差异不显著,茶树花粉离体萌发最佳培养基为150g/L蔗糖+150mg/L硼酸+100mg/L硝酸钙。离体萌发法和TTC染色法测定茶树花粉活力进行比较,2种方法得到茶树花粉活力最佳分别为90.04%、83.17%,且差异不显著。因此,花粉离体萌发法和TTC染色法都可用于测定茶树花粉活力,10g/L TTC用于快速检测茶树花粉活力效果最佳。Pollens from Camellia sinensis cv.‘Fuding-dabaicha’were selected as the experimental materials in this study.Taking the concentration of the sucrose,the boric acid and the calcium nitrate into consideration,we carried out the orthogonal test to optimize germination medium of the tea tree pollen in vitro.The in vitro germination and TTC staining of the pollen were compared to investigate the method for rapid detecting of pollen vigor in C.sinensis.The results showed that boric acid was the main factor affecting the germination of tea pollen in vitro,and the concentration of the sucrose had no significant effect on the pollen germination in vitro.The optimized germination medium in vitro for the pollen contained 150 g/L sucrose,150 mg/L boric acid and 100 mg/L calcium nitrate;We compared the pollen vigor measured by TTC staining and in vitro germination,respectively.The pollen vigor was 90.04%(in vitro germination)and 83.17%(TTC staining),respectively.And the detection method had no significant effect on the pollen vigor.Therefore,TTC staining and in vitro germination could be used to detect the pollen vigor in C.sinensis.,and the TTC staining,with 10 g/L,was a rapid method to test the pollen vigor in the tea plant.
