摘 要:目的:探究环状RNA(circR NA) hsa_circ_0001785在乳腺癌组织和细胞中的表达变化及其对乳腺癌细胞增殖、迁移和侵袭能力的影响。方法:qRT-PCR实验检测hsa_circ_0001785在乳腺癌组织和乳腺癌细胞(MDA-MB-231和SK-BP-3)中的相对表达水平; CCK-8和克隆形成实验检测沉默或上调表达hsa_circ_0001785对MDA-MB-231细胞活性和克隆形成能力的影响;划痕愈合实验和Transwell侵袭实验检测沉默或上调表达hsa_circ_0001785对MDA-MB-231细胞迁移及侵袭能力的影响。结果:hsa_circ_0001785在乳腺癌组织中的相对表达水平明显高于癌旁组织,hsa_circ_0001785在MDA-MB-231和SK-BP-3细胞中的相对表达水平明显高于人乳腺上皮细胞MCF10A。在MDA-MB-231细胞沉默hsa_circ_0001785,MDAMB-231细胞的活性和克隆形成能力明显降低,迁移距离显著减少,侵袭能力也明显下降。而在MDA-MB-231细胞中上调表达hsa_circ_0001785,MDA-MB-231细胞的活性和克隆形成能力显著升高,迁移距离明显升高,侵袭能力也明显升高。结论:hsa_circ_0001785在乳腺癌组织和乳腺癌细胞(MDA-MB-231和SK-BP-3)中的表达水平明显升高;沉默hsa_circ_0001785显著抑制乳腺癌MDA-MB-231细胞的增殖、迁移和侵袭能力,而上调表达hsa_circ_0001785明显促进乳腺癌MDA-MB-231细胞的增殖、迁移和侵袭能力。Objective:To explore the expression of the circular RNA(circRNA)hsa-circ_0001785in breast cancer tissues and cell lines,and to study the effect of hsa_circ_0001785knockdown on the proliferation,migration and invasion of breast cancer cells.Methods:The expression levels of hsa_circ_0001785in breast cancer tissues and breast cancer cell lines(MDA-MB-231and SK-BP-3)were measured by quantitative real-time PCR(qRT-PCR).To assess the effect of hsa_circ_0001785on the viability and proliferation of breast cancer cells,CCK-8and colony formation assays were performed in MDA-MB-231cells.Transwell migration and wound scratch assays were carried out to evaluate the effect of hsa_circ_0001785on the migration and invasion ability of MDA-MB-231cells.Results:The expression levels of hsa_circ_0001785were higher in breast cancer tissues and MDA-MB-231and SK-BP3cells than in adjacent normal tissues and human mammary epithelial cells(MCF10A).Knockdown of hsa_circ_0001785markedly repressed the proliferation migration and invasion of MDA-MB-231cells.Besides upreg-ulation of hsa_circ_0001785promoted the proliferation,migration and invasion of MDA-MB-231cells.Conclu-sion:Knockdown of hsa_circ_0001785remarkably represses the proliferation,migration and invasion of MDA-MB-231cells.
