摘　　要：目的探讨肿瘤抑制基因1(metastasis suppres-sor 1,MTSS1)在宫颈鳞癌及子宫内膜腺癌细胞中的表达。方法选取30例宫颈鳞癌患者病理标本作为研究组1,同时选取30例经宫颈液基细胞学证实无异常的宫颈病理标本作为对照组1。另选取30例子宫内膜腺癌患者病理标本作为研究组2,同时选取30例子宫平滑肌瘤患者正常子宫内膜病理标本作为对照组2。通过免疫组织化学方法和实时荧光定量PCR(q-PCR)测引物序列及Western蛋白印迹法分析MTSS1在宫颈鳞癌及子宫内膜腺癌中的表达及其临床病理因素。结果运用q-PCR测引物序列及Western蛋白印迹法检测表明研究组1和2中MTSS1阳性表达率均明显高于相应对照组,差异具有统计学意义(P <0.05);免疫组化法检测表明研究组1和2中MTSS1阳性表达率均明显高于相应对照组;组间对比差异具有统计学意义(P <0.05)。结论两种方法检测结果相同,均证明MTSS1在宫颈鳞癌及子宫内膜腺癌中的高表达,进一步在蛋白水平证实MTSS1基因的表达可能与妇科恶性肿瘤的发生发展密切相关,为临床癌症筛查提供了诊断方法。Objective To investigate the expression of metastasis suppressor1(MTSS1)in cervical squamous cell carcinoma and endometrial adenocarcinoma cells.Methods Pathological specimens of30patients with cervical squamous cell carcinoma were selected as study group1,and30cervical pathological specimens confirmed by cervical liquidbased cytology were selected as control group1.Another30cases of endometrial adenocarcinoma were selected as study group2,while30cases of normal endometrial pathological specimens of patients with uterine leiomyoma were selected as control group2.The expression of MTSS1in cervical squamous cell carcinoma and endometrial adenocarcinoma and its clinicopathological factors were analyzed by immunohistochemical84China Continuing Medical Education,Vol.11,No.1method,real-time fluorescence quantitative PCR(q-PCR)primer sequence and Western blotting.Results The positive expression rates of MTSS1in the study group1and2were significantly higher than those in the control group by using q-PCR primer sequence and Western blotting(P<0.05).Immunohistochemical assay showed that MTSS1positive expression rates in study group1and2were significantly higher than those in control group,and there was significant difference between the two groups(P<0.05).Conclusion The results of the two methods are the same,which proves that MTSS1is highly expressed in cervical squamous cell carcinoma and endometrial adenocarcinoma.Further,the protein level confirms that MTSS1gene expression may be closely related to the occurrence and development of gynecological malignant tumors,which provides a diagnostic method for clinical cancer screening.
