摘 要:目的观察膀胱癌组织中微小RNA212(miR-212)的表达及上调miR-212表达对膀胱癌细胞增殖、迁移及侵袭的影响,探讨miR-212是否通过调控高迁移率族蛋白2(HMGA2)来实现其作用。方法取31例膀胱癌患者的膀胱癌病理组织标本及配对癌旁正常组织,采用逆转录聚合酶链反应(qRT-PCR)检测膀胱癌组织miR-212mRNA表达。选择人膀胱癌T24细胞,分为miR-212组(转染miR-212 mimics)和对照组(转染空白的miR-212NC),培养48 h后,采用MTT法检测膀胱癌细胞增殖能力,采用Transwell实验测定膀胱癌细胞迁移和侵袭能力,采用Western blotting法检测膀胱癌细胞HMGA2蛋白表达。采用荧光素酶报告实验验证HMGA2是否是miR-212的下游靶基因。结果膀胱癌组织中miR-212 mRNA表达低于癌旁正常组织(P <0. 05)。miR-212组细胞增殖、迁移和侵袭能力均低于对照组(P均<0. 05),miR-212组HMGA2蛋白表达低于对照组(P <0. 05)。在线数据库及荧光素酶报告实验预测并验证HMGA2是miR-212的下游靶基因。结论膀胱癌组织中miR-212表达降低,上调miR-212表达可抑制膀胱癌细胞的增殖、迁移和侵袭能力,其作用与HMGA2密切相关。Objective To observe the expression of microRNA212(miR-212)in the bladder cancer tissues and its effects of up-regulation on proliferation,migration,and invasion of bladder cancer cells,and to explore whether miR-212can achieve its effects by regulating high-mobility group box2(HMGA2).Methods The qRT-PCR method was used to detect the expression level of miR-212in31cases of bladder cancer tissues and adjacent normal tissues.Human bladder cancer T24cells were divided into two groups:miR-212group(transfected with miR-212mimics)and the control group(transfected with blank miR-212NC).After48hours of incubation,MTT method and Transwell experiment was used to observe the cell proliferation,invasion and migration.Western blotting was used to detect the protein expression level of HMGA2.Luciferase reporter experiment was used to verify whether HMGA2was the downstream target gene of miR-212.Results Compared with the normal tissues,the expression of miR-212in the bladder cancer tissues decreased significantly(P<0.05).The proliferation,migration and invasion of the miR-212group were lower than those of the control group(all P<0.05).The expression of HMGA2protein in the miR-212group was lower than that of the control group(P<0.05).Online database and luciferase reporter experiments predicted and verified that HMGA2was the downstream target gene of miR-212.Conclusion The expression of miR-212is decreased in the bladder cancer tissues,and up-regulation of miR-212expression can inhibit the proliferation,migration and invasion of bladder cancer cells,and its effect is closely related to HMGA2.
