摘 要:目的探讨高迁移率族蛋白B1(HMGB1)对人牙周膜成纤维细胞(PDLCs)骨钙蛋白(OC)和骨涎蛋白(BSP)表达影响。方法原代培养人PDLCs,取传4~6代细胞,随机分为HMGB1组、对照组,HMGB1组给予100ng/mL HMGB1处理,对照组不予HMGB1处理。采用RT-PCR法和Western blotting法分别检测各组OC、BSP mRNA和蛋白表达。结果 HMGB1组OC mRNA和蛋白相对表达量明显高于对照组,BSP mRNA和蛋白相对表达量明显低于对照组(P均<0. 05)。结论 HMGB1可能通过影响人PDLCs中OC和BSP表达,从而参与牙周病中牙槽骨破坏,继而促进牙周病发展。Objective To investigate the influence of high mobility group box1(HMGB1)on the expression of osteocalcin(OC)and bone sialoprotein(BSP)of human periodontal ligament fibroblast cells(PDLCs).Methods PDLCs were collected from original generation of tissue cultivation and the cells in generation4-6were used in the experiment.The cells were randomly divided into the HMGB1group and control group.The cells in the HMGB1group were stimulated with HMGB1at concentrations of100ng/mL,but in control group without HMGB1.RT-PCR and Western blotting were performed to check the expression of OC,BSP mRNA and protein in each group.Results Compared with the control group,OC increased at both mRNA and protein levels in the HMGB1group(both P<0.05).However,the mRNA and protein expression levels of BSP were significantly lower in the HMGB1group than in the control group(both P<0.05).Conclusion By influencing OC and BSP expression in PDLCs,HMGB1can participate in the destruction of the alveolar bone in periodontal disease and promote the development of periodontal disease.
