摘  要:研究动态超高压微射流技术对蔓三七叶黄酮得率的影响。以单因素实验为基础,选取乙醇浓度、提取温度、提取时间、动态超高压微射流均质压力为主要因素进行研究,通过响应面优化分析,优化蔓三七叶黄酮的提取工艺,并对其体外抗氧化活性进行评价。结果表明,最佳提取工艺条件为:乙醇浓度68.0%、提取时间68 min和均质压力80 MPa,在此条件下黄酮得率为2.08%。在质量浓度0.2~1.0 mg/mL范围内,蔓三七叶黄酮体外抗氧化活性呈现出量效关系趋势。在浓度为0.8 mg/mL时,蔓三七叶黄酮的总抗氧化能力值为(11.25±0.45)U/mL,为同等浓度下抗坏血酸的总抗氧化能力[(13.56±0.50)U/mL]的90%以上,各浓度蔓三七叶黄酮对DPPH·和·OH清除能力均有较强的清除能力,是一种良好的天然抗氧化剂。The effect of dynamic high-pressure microfluidization treatment on the yield of Gynura procumbens leaves flavonoids were studied in this paper.Based on the experiment of single factor and the response surface methodology were used to optimize the extraction processing parameters and investigate the effects of the following four factors on extraction rate of flavonoids:dynamic high-pressure microfluidization retreatment pressure,ethanol concentration,extraction temperature and extraction time.The optimal extraction processing parameters were aqueous ethanol concentration 68.0%,extraction time 68min,microfluidization pressure 80.0MPa,Under these conditions the yield of flavonoids was 2.08%.At the mass concentration of 0.2~1.0mg/mL,the total antioxidant activity was(11.25±0.45)U/mL and reached more than that of 90% Vc with the same mass concentration.The Gynura procumbens leaves flavonoids of different concentrations have strong scavenging capacity for DPPH and OH scavenging ability.It was a good natural antioxidant.
