摘  要:从武汉华夏理工学院一食堂附近取土壤样品,分离土壤微生物,筛选能产生淀粉酶的菌种,并进行初步鉴定。经紫外线诱变,采用碘淀粉显色法进行初筛,得到一株产酶量高、性状优良的突变菌株。通过研究不同p H值、装液量、碳源条件下淀粉酶的活性和运用正交实验得出最佳培养基的配方,初步确立高产菌株的最佳发酵条件,同时对菌株产生的酶进行初步提取,将得到的粗酶进行酶活力的测定。结果表明,本试验中筛选得到的淀粉酶其酶活性提高了23.7%。Soil samples were taken from acanteen of Wuhan Huaxia University of Technology.Through the isolationof soil microorganisms,screening of amylase-producing strains,preliminary identification and UV mutagenesis,amutant strain with high enzyme production and excellent properties was obtained by primary screening with iodinestarch chromogenic method.By studying the amylase activity under different pH value,liquid loading and thecarbon source conditions and using orthogonal experiment to obtain the optimal culture medium formula,the optimalfermentation conditions for high-yield strains were preliminarily established.and the enzyme produced by the strainswas preliminarily extracted,and the enzyme activity of the crude enzyme obtained was measured.The results showedthat the enzyme activity of the amylase screened in this experiment increased by23.7%.
