摘　　要：目的探讨拓扑异构酶2a(TOP2a)、丝/苏氨酸激酶NIMA相关激酶2(Nek2)、核苷酸还原酶亚基1(RRM1)在胃癌癌组织中的表达及临床意义。方法回顾性研究,通过免疫组化法测定60例胃癌癌组织(观察组)、30例正常组织(对照组) Top2A、NEK2、RRM1表达,并分析其表达与胃癌患者临床病理特征的关系。结果观察组Top2A、NEK2、RRM1阳性率分别为66. 67%、65. 00%、83. 33%,均显著高于对照组的10. 00%、13. 33%、16. 67%,差异有统计学意义(P <0. 05);胃癌组织Top2A表达与患者TNM分期、分化程度、浸润深度及淋巴结转移显著相关(P <0. 05),NEK2表达与患者分化程度、淋巴结转移显著相关(P <0. 05),RRM1表达与患者分化程度、浸润深度显著相关(P <0. 05);胃癌组织Top2A表达与NEK2、RRM1均呈正相关(P <0. 05)。结论胃癌癌组织中TOP2a、Nek2及RRM1高表达,且三者正相关,与肿瘤分化程度、浸润深度或淋巴结转移密切相关,检测TOP2a、Nek2及RRM1有利于肿瘤发生、进展评估,指导临床治疗。Objective To investigate the expression and clinical significance of topoisomerase 2a(TOP2a),silk/threonine kinase NIMA-related kinase 2(Nek2)and nucleotide reductase subunit 1(RRM1)in gastric cancer.Methods The expressions of Top2A,NEK2 and RRM1 in 60 gastric cancer tissues(observation group)and 30 normal tissues(control group)were determined by immunohistochemistry,the relationship between the expression and the clinicopathological features of gastric cancer patients was analyzed.Results The positive rates of Top2A,NEK2 and RRM1 in the observation group were 66.67%,65.00% and 83.33%,respectively,which were significantly higher than those in the control group(10.00%,13.33%,16.67%)(P<0.05).The expression of Top2A was significantly correlated with TNM stage,differentiation,depth of invasion and lymph node metastasis(P<0.05).NEK2 expression was significantly correlated with patient differentiation and lymph node metastasis(P<0.05).RRM1 expression was significantly correlated with patient differentiation and infiltration depth(P<0.05).Top2A expression in gastric cancer tissues was correlated with NEK2 and RRM1(P<0.05).Conclusion TOP2a,Nek2 and RRM1 are highly expressed in gastric cancer tissues and positively correlated.They are closely related to tumor differentiation,depth of invasion or lymph node metastasis.The detection is beneficial to tumorigenesis,progression evaluation and the guidance to the clinical treatment.
