摘 要:目的改良并建立C57BL/6近交系小鼠前列腺癌原位与皮下种植瘤模型,结合ZYX-1-9的药理学特性及动物模型的生物学特性,评价该药的治疗效果,比较腹腔用药与瘤内用药的疗效差异。方法建立小鼠前列腺原位癌模型40只、皮下肿瘤模型80只,9 mg/kg、6 mg/kg、3 mg/kg ZYX-1-9的3种剂量治疗,同时设置阴性对照组,原位癌采用腹腔注射用药,皮下肿瘤采用瘤内注射和腹腔注射用药。观察肿瘤转移情况、计算抑瘤率、HE染色观察。结果成功建立模型。对40只原位癌模型行腹腔注射用药、80只皮下肿瘤模型行瘤内注射用药和腹腔注射用药治疗后,ZYX-1-9的抑瘤率较高。小鼠前列腺癌皮下肿瘤的瘤内注射法抑瘤率高于腹腔注射法。结论 ZYX-1-9抑瘤作用显著,对前列腺癌具有良好的治疗效果。ZYX-1-9对前列腺癌行瘤内注射用药,疗效显著。Objective The C57BL/6 inbred mouse prostate cancer in situ and subcutaneous implant tumor model was modified and established.The pharmacological properties of ZYX-1-9 and the biological characteristics of the animal model were combined to evaluate the therapeutic effect of the drug.The difference in efficacy of intratumoral administration.Methods A total of 40 mice with prostate cancer in situ and 80 subcutaneous tumor models were established,9 mg/kg,6 mg/kg,and 3 mg/kg ZYX-1-9.In situ carcinoma was administered intraperitoneally,and subcutaneous tumors were administered intratumorally and intraperitoneally.The tumor metastasis was observed,the tumor inhibition rate was calculated,and HE staining was observed.Results Successfully built the model.ZYX-1-9 has a higher anti-tumor rate after intraperitoneal injection of 40 in situ carcinoma models and 80 subcutaneous tumor models by intratumoral injection and intraperitoneal injection.The tumor inhibition rate of intratumoral injection of mouse prostate cancer subcutaneous tumor is higher than that of intraperitoneal injection.Conclusion ZYX-1-9 has a significant anti-tumor rate and has a good therapeutic effect on prostate cancer.ZYX-1-9 has a significant effect on intratumoral injection of prostate cancer.
