摘 要:目的:优化酶法辅助提取民族药地板藤总多糖的工艺条件。方法:在单因素试验基础上,采用Box-BehnkenDesign-响应面法,以酶用量、酶解时间、提取料液比为响应因子,以地板藤总多糖的提取率为响应值,采用3因素3水平的BBD设计进行试验,研究各因素及其交互作用对地板藤中总多糖提取率的影响。结果:响应面分析法优化的最佳提取工艺为酶用量为1.5%、酶解时间63.31min、提取料液比1:32.92,预测最高提取率9.23%;验证试验中地板藤总多糖的提取率为8.51%(RSD=0.93%,n=5)。结论:酶法辅助提取地板藤总多糖的提取率较高,工艺合理、简便,可用作地板藤多糖的提取。Objective:To optimize the enzymatic extraction of total polysaccharides from Ficus tikoua Bur.Methods:The process was optimized by Box-behnken-response surface methodology based on the single factor tests.By using enzyme dosage,enzyme time and feed liquid ratio as response factors,total polysaccharides used as response value,3-factors and 3-level analysis was adopted for the experiment.Results:The optimal extraction technology was as enzyme dosage of 1.5%,enzyme time of 63.31 min and feed liquid ratio of 1:32.92,and the predicted highest extraction yield was 9.23 mg/g.In validation test,the extraction rate of total polysaccharides was 8.51%(RSD=0.93%,n=5).Conclusion:The optimized extraction technology is effective,reasonable and simple,and can be used for the extraction of total polysaccharides from Ficus tikoua Bur.
