NDRG1及NDRG2基因在结直肠癌组织中的表达及临床意义

摘　　要：目的:探讨NDRG1及NDRG2基因在结直肠癌中的表达与临床病理特征及预后的关系。方法:采用免疫组织化学检测92例结直肠癌组织及其配对癌旁组织中NDRG1及NDRG2蛋白的表达情况,统计学分析其与结直肠癌临床病理特征及预后的关系。结果:(1) NRDG 1及NDRG 2在92例结直肠癌组织中均有不同程度的表达:NRDG 1的阳性率为20.7%,NDRG 2的阳性率为23.9%,均明显低于其配对癌旁正常组织,差异有统计学意义(P<0.01)。(2)肿瘤浸润程度高(T3、T4)的结直肠癌组的NDRG1阳性率明显低于肿瘤浸润程度低(T1、T2)的结直肠癌组的阳性率(P<0.05),差异有统计学意义。其阳性表达与病人的年龄、性别、肿瘤的分化程度、分期及有无淋巴结转移无关(P>0.05);有淋巴结转移的结直肠癌组的NDRG2的阳性率明显低于无淋巴结转移的结直肠癌组的阳性率(P<0.05),差异有统计学意义;其阳性表达与病人的年龄、性别,肿瘤的分期、分化程度、浸润程度无关(P>0.05)。(3) NDRG 1及NDRG 2表达水平与结直肠癌病人3 a生存率及5 a生存率无明显相关,无统计学意义。(4)结直肠癌中NDRG1及NDRG2无明显相关性(P>0.05)。结论:NDRG 1及NDRG2基因在结直肠癌的发展、侵袭及转移过程中起到重要作用,对临床诊断结直肠癌病情发展程度具有一定的参考意义。Objective:To explore the expression of tumor-related genes of NDRG1 and NDRG2 in colorectal adenocarcinomas and the relationship with clinical pathological features and prognosis. Methods: Immunohistochemical two-staged methods were used to detected NDRG1 and NDRG2 in 92 cases of colorectal adenocarcinomas and cases of colorectal adenocarcinomas accompanying with lymph node metastasis. Results:(1)The expression of NDRG1 and NDRG2 in colorectal adenocarcinomas was positive of different levels:The positive expression of NDRG1 is 20.7 %(19/92),The positive expression of NDRG2 is 23.9 %(22/92), both remarkably lower than that in oncopathogenesis( P < 0.01 ).(2)For NDRG1,The positive expression of higher invasion(T3、T4)in colorectal adenocarcinomas was remarkably lower than that of low invasion(T1、T2)( P < 0.05 ).There was no significant difference among different ages,differentiation degrees,genders,stages or lymphatic invasion groups for NDRG1 expression( P > 0.05 ).The positive rate of NDRG2 accompanying with lymph node metastasis in colorectal adenocarcinomas was remarkably lower than that accompanying without lymph node metastasis( P < 0.05 ).There was no significant TNM stages,ages,differentiation degrees,genders or lymphatic invasion difference among different groups( P > 0.05 ).(3)The expression of NDRG1 and NDRG2 had no statistical significance for three and five year survival of patients.(4)There was no remarkably correlation between NDRG1 and NDRG2 in colorectal adenocarcinomas( P > 0.05 ). Conclusion: NDRG 1 and NDRG2 play an important part in the progress of tumor development,invasion or metastasis.It provides the the guiding significance for the clinical diagnosis.
