摘 要:目的探讨黄嘌呤氧化酶活性对大鼠血清胰岛素水平的影响。方法 2017年9月以39只雄性SD大鼠为研究对象,按体重分层随机分为空白对照组(C组)、酵母组(Y组)和"酵母+苯溴马隆"组(YB组),每组13只。Y组给予酵母粉[10 g/(kg·d)],YB组给予酵母粉[10 g/(kg·d)]和苯溴马隆[5 mg/(kg·d)],C组给予等体积蒸馏水,每天分2次灌胃给药,持续4周。实验前和实验后,大鼠外眦静脉取血,测定大鼠血清中尿酸(UA)、黄嘌呤氧化酶(XO)、葡萄糖、胰岛素(INS)水平,并计算胰岛素抵抗指数。结果干预后,Y组大鼠血清UA水平没有明显升高(t=-1.169,P>0.05)。Y组大鼠血清XO活性升高[干预前(9.7±0.8) U/L,干预后(10.8±1.0) U/L,t=-3.700,P<0.05],且高于对照组[(9.0±1.1) U/L,t=-4.384,P<0.05];同时Y组大鼠血清INS水平降低[干预前(1.61±0.15) m U/L,干预后(0.91±0.29) mU/L,t=-8.381,P<0.05],并低于对照组[(1.15±0.21) m U/L,t=2.970,P<0.05]。结论黄嘌呤氧化酶活性升高使大鼠血清胰岛素水平降低,可能是胰岛B细胞分泌功能损伤所致。Objective To study the effect of xanthine oxidase activity on the serum insulin level of rats.Methods 39 cases of male SD rats in September 2017 were selected as the research objects and randomly divided into the control C group,yeast Y group and yeast and benzbromarone YB group,with 13 cases in each,the Y group were given yeast powder 10 g/(kg·d),the YB group were given yeast powder 10 g/(kg·d)and benzbromarone 5 mg/(kg·d),the group C were given an equal volume of distilled water,for intragastric administration twice per day,for consecutive four weeks,and the rats were for intravenous blood before and after experiment,and the UA,XO,GLUOCSE,INS level of rats were measured,and the insulin resistant index was calculated.Results After intervention,the serum UA level in the Y group had no obvious increase(t=-1.169,P>0.05),and the serum XO activity in the Y group increased(before intervention(9.7±0.8)U/L,after intervention(10.8±1.0)U/L(t=-3.700,P<0.05),higher than that in the control group(9.0±1.1)U/L(t=-4.384,P<0.05);at the same time,the serum INS level decreased in the Y group(before intervention(1.61±0.15)mU/L,after intervention(0.91±0.29)mU/L,(t=-8.381,P<0.05),lower than that in the control group(1.15±0.21)mU/L(t=2.970,P<0.05).Conclusion The increase of xanthine oxidase activity can decrease the serum insulin level of rats,which may be caused by pancreatic B cell secretory function injury.
