摘 要:笔者采用酶解法改进三公膏传统生产工艺和采用双缩脲法测定三公膏中多肽含量。结果表明,在三公膏传统工艺基础上增加酶解、离心除杂、脱油等步骤,使生产周期从5天4夜缩短到2天左右,出膏率由3. 0%左右提高到8. 84%,多肽含量由11. 90%提高到40. 36%;双缩脲法测定多肽含量简便、准确。Sangonggao is a popular traditional healthcare medicine prepared by the Hainan Li Ethnic Group,and its preparation is time consuming and less effective.An enzymatic hydrolysis was used to improve the traditional preparation process of Sangonggao,and the polypeptide content of Sangonggao was determined by employing the Coomassie blue staining method or the biuret method.The results showed that the preparation process of Sangongao was improved through additional steps such as glycolysis,centrifugation and removal of fats and oils in the basis of the traditional preparation process.With this improved method the preparation was shortened from 5 days and 4 nights to about 2 days and the Sangonggao paste yield increased from about 3.0%to 8.84%;the polypeptide content of the prepared Sangonggao was raised from 11.90%to 40.36%.The biuret method was found to be simple and accurate in determination of polypeptide content in Sangonggao.
