苏丹红I对大鼠CYP 2E1、CYP 3A1酶活性、CYP 450含量及血液成分的影响

摘　　要：【目的】为探讨苏丹红I(Sudan I)对大鼠肝脏细胞色素P450酶系CYP 2E1和CYP 3A1活性的影响及血液指标的变化。【方法】将30只清洁级SD大鼠随机分为2组:对照组和Sudan I处理组(包括4个剂量处理组),连续饲喂10 d,采集肝脏和血液作为检测样品,运用酶活性测定技术和血液成分进行分析。【结果】Sudan I能够诱导CYP 2E1、CYP 3A1酶活性及CYP 450s表达增加(P <0. 05,P <0. 01),使红细胞数和血红蛋白的量表现为减少,白细胞数和血小板的量表现为增加。【结论】Sudan I能导致大鼠CYP 2E1、CYP 3A1酶活性、CYP 450含量及血液成分的变化,引发毒性效应导致肝脏受损。【Objective】The study aimed to explore the effect of Sudan I on biochemical indicator of the CYP 2E1,CYP 3A1 and CYP 450 content in liver microsome,and the concentration of hematology index in rats.【Method】30 SD rats were randomly divided into two groups:control group and Sudan Itreatment group(There are four doses of the treatment group).All rats were killed and their liver and blood were collected for test 10 days after treatment.Measure the enzyme activity and blood components to analyze the change.【Result】The experiment results were indicated that the activity of CYP 450s,CYP 2E1,CYP 3A1 in rat liver microsome(P<0.05,P<0.01);The red blood cell and hemoglobin were decreased and the white blood and platelet were increased in Sudan Igroup.【Conclusion】Sudan I could educed the change in the activity of CYP 2E1,CYP 3A1,CYP 450 content and blood components in rats,then leaded to the poisoning of organism and injury of rat liver.
