摘 要:目的:探讨结直肠癌患者基质金属蛋白酶-26(MMP-26) mRNA和蛋白质的表达及其与临床病理的关系。方法:采用RT-PCR及Western Blot法检测MMP-26在31份结直肠癌及31份癌旁正常组织样本中的表达水平并比较,采用免疫组化Max Vision法检测74份结直肠癌和65份癌旁正常组织中MMP-26的表达情况并比较,分析MMP-26表达水平与临床病理的关系。结果:31份结直肠癌样本中,有20份(64.52%)样本的MMP-26 mRNA表达明显高于癌旁正常组织,有21份(67.74%)MMP-26蛋白质表达水平明显高于癌旁正常组织。免疫组化结果显示,MMP-26在癌组织中阳性表达率为63.51%(47/74),明显高于癌旁正常组织26.15%(17/65),差异有统计学意义(P<0.05);MMP-26蛋白质表达水平与肿瘤浸润深度、淋巴结转移及TNM分期有关(P<0.05),与患者的性别、年龄、肿瘤大小及分化程度均无相关性。结论:MMP-26在结直肠癌组织中呈高表达,且与临床病理特征相关,其可能在癌细胞浸润转移中发挥促进作用,可作为判断结直肠癌进展及预后的重要标记物。Objective: To investigate expressions of matrix metalloproteinase-26(MMP-26) and protein in colorectal cancer and its relationship with clinicopathological features. Methods: The expression levels of MMP-26 in 31 colorectal cancers and 31 adjacent normal tissues were detected by using RT-PCR and Western Blot and compared. Then, the expression levels of MMP-26 in 74 colorectal cancers and 65 adjacent normal tissues were detected by using the immunohistochemical MaxVision method. Further, the relationship among MMP-26 expression level and clinicopathological features was analyzed. Results: Among the 31 colorectal cancer samples, 20(64.52%) samples had signifi cantly higher MMP-26 mRNA expression than the adjacent normal tissues, and 21(67.74%) samples had signifi cantly higher MMP-26 protein expression than the adjacent normal tissues. The immunohistochemical analysis showed that the positive expression rate of MMP-26 in the cancer tissues was 63.51%(47/74), which was signifi cantly higher than that in the adjacent normal tissues(26.15%, 17/65), and the difference was statistically signifi cant(P<0.05). The expression level of MMP-26 protein was correlated with tumor invasion depth, lymph node metastasis and TNM staging(P<0.05), but was not correlated with gender, age, tumor size and differentiation degree. Conclusions: MMP-26 is highly expressed in the colorectal cancer tissues and is correlated with the clinicopathological features. Moreover, it may play a role in the cancer cell invasion and metastasis, and may be an important marker for judging the progression and prognosis of colorectal cancer.
