摘 要:(1)目的探讨二甲双胍对TPC-1人乳头瘤状甲状腺癌细胞、Eca-109人食管癌细胞、SHSY5Y人神经母细胞瘤细胞、PC-3人前列腺癌细胞、HT-29人结肠癌细胞及HCT-8人结肠癌细胞增殖能力的影响,分析二甲双胍的抗肿瘤作用。(2)方法将6种恶性肿瘤细胞均匀地种植于96孔板内,37℃、双气、饱和湿度下常规培养;MTT实验检测各肿瘤细胞在不同浓度盐酸二甲双胍作用48小时的细胞增殖能力及1.5mg/mL盐酸二甲双胍作用24小时、48小时、72小时的细胞增殖能力。(3)结果盐酸二甲双胍抑制了TPC-1人乳头瘤状甲状腺癌细胞、Eca-109人食管癌细胞、SH-SY5Y人神经母细胞瘤细胞、PC-3人前列腺癌细胞、HT-29人结肠癌细胞及HCT-8人结肠癌细胞的增殖(P <0.05),且呈剂量及时间依赖性。(4)结论二甲双胍可降低实验中各种肿瘤细胞的增殖能力,抑制细胞增殖,提示二甲双胍可能具有广泛的抗肿瘤作用。Objective To explore the influence of MET on the proliferation of some malignant tumor cells about TPC-1 human papillary thyroid cancer cells,Eca-109 human esophageal cancer cells,SH-SY5Y human neuroblastoma cells,PC-3 human prostate cancer cells,HT-29 human colon cancer cells and HCT-8 human colon cancer cells,and analyze the anti-tumor effect of MET.Methods Six kinds of malignant tumor cells were cultivatedin 96-well plates evenly and culture cells in 37℃ and double gas and saturated humidity.The proliferation of malignant tumor cells were determined by MTT experiment testing in different concentrations of MET and in different time with 1.5mg/mL MET.Results MET inhibited the proliferation of malignant tumor cells about TPC-1 human papillary thyroid cancer cells,Eca-109 human esophageal cancer cells,SH-SY5Y human neuroblastoma cells,PC-3 human prostate cancer cells,HT-29 human colon cancer cells and HCT-8 human colon cancer cells in dose and time(P<0.05).Conclusion MET can reduce the proliferation of various malignant tumor cells in the experiment,suggesting that MET may have a broad anti-tumor effect.
