摘 要:目的探讨微小RNA-150(miR-150)在非小细胞肺癌(NSCLC)组织中的表达情况及其临床意义。方法收集本院2014年9月至2017年6月手术切除的NSCLC患者癌组织及配对癌旁组织73对,采用实时定量PCR(QPCR)检测NSCLC组织的miR-150水平,分析其与NSCLC临床病理特征(性别、年龄、吸烟史、肿瘤大小、淋巴结转移、TNM分期和病理类型)的关系,根据随访资料分析不同miR-150水平NSCLC患者的预后情况。结果 QPCR结果显示NSCLC组织中的miR-150水平为2. 014±0. 227,高于癌旁组织的1. 023±0. 154,差异有统计学意义(P<0. 05); NSCLC患者的miR-150表达与性别、年龄、吸烟史和病理类型均无关,而与淋巴结转移、肿瘤大小和TNM分期有关(P<0. 05),且淋巴结转移阳性、肿瘤较大及临床分期较晚患者的miR-150水平较高。Kaplan-Meier生存曲线分析结果显示,miR-150高表达者的中位生存期为27. 0个月,少于低表达者的34. 0个月(P<0. 05)。结论 MiR-150在NSCLC组织中高表达并参与了NSCLC的发生发展,该miRNA水平较高者的预后较差,可作为NSCLC潜在的分子诊断及预后监测标志物。Objective To evaluate the expression and clinical significance of microRNA-150( miR-150) in non-small cell lung cancer( NSCLC) tissues. Methods Seventy-three pairs of cancerous tissues and matched adjacent tissues from 73 patients with NSCLC who underwent surgical excision from September 2014 to June 2017 were collected. Quantitative real-time PCR( QPCR) was performed to evaluate miR-150 level of the above tissues. The relationship between NSCLC and its clinicopathological features( sex,age, smoking history,tumor size,lymph node metastasis,TNM stage and pathological type) was analyzed. The prognosis of NSCLC patients with different miR-150 levels was analyzed based on follow-up data. Results The level of miR-150 in NSCLC tissues was2. 014±0. 227,higher than 1. 023±0. 154 in adjacent tissues( P<0. 05). The expression of miR-150 in NSCLC patients was not related to gender,age,smoking history and pathological type,but related to lymph node metastasis,tumor size and TNM stage( P< 0. 05).The miR-150 level was higher in patients with positive lymph node metastasis,larger tumor size and late clinical stage. Kaplan-Meier survival curve analysis showed that the overall survival of patients with high-expression of miR-150 was 27. 0 months,which was lower than 34. 0 months of patients with low-expression( P<0. 05). Conclusion MiR-150 is highly expressed in NSCLC tissues and participates in the occurrence and development of NSCLC. Those with higher miR-150 level have poor prognosis and miR-150 can be used as potential diagnostic and prognostic monitoring markers of NSCLC.
