摘　　要：骨肉瘤发病机制极其复杂,目前尚未明确。研究表明胰岛素样生长因子家族(IGFs)在骨肉瘤的发生发展中有重要作用。IGF-1、IGF-2高表达与骨肉瘤的发生呈正相关,IGF-IR相关的异常信号通路参与骨肉瘤的发生和发展,IGF-IR也与骨肉瘤的不良预后密切相关,IGF-1R表达水平越高,提示预后越差。IGF家族中其它成员如IGF-2R、IGFBP等均被证实参与了骨肉瘤的发生发展。近年来,以IGF-1R为主的靶向治疗在骨肉瘤辅助治疗中初见成效,各期临床试验正在稳步进行,但确切的临床疗效还有赖于对IGFs和骨肉瘤的更进一步研究和探索。本文对IGFs在骨肉瘤的发生发展及治疗中的相关作用进行综述。The pathogenesis of osteosarcoma is extremely complex and still remains unclear.It has been shown that IGFs play an important role in the development of osteosarcoma.The high expression of IGF-1 and IGF-2 are positively correlated with the occurrence of the tumor.IGF-IR related signaling pathways are also involved in the development of osteosarcoma.It is suggested that the poor prognosis may be associated with IGF-1R,the higher the expression of IGF-1R is,the worse the prognosis of the tumor is.Other members of the IGF family,such as IGF-2R and IGFBP,have also been proved to contribute to the tumor’s growth.In recent years,targeted therapy based on IGF-1R has achieved initial results in the adjuvant treatment of osteosarcoma.However,the exact clinical effect depends on the further study and exploration of osteosarcoma and IGFs.This article reviews the role of IGFs in the development and treatment of osteosarcoma.
