摘  要:γ-氨基丁酸(Gamma-aminobutyric acid,GABA)在食品和医药领域具有重要作用。许多方法已用于测定GABA,包括以Cu2+为稳定剂的预染纸色谱-分光光度法。本论文报道了一种新的GABA定量方法:无Cu2+洗脱预染纸色谱-分光光度法(No-Cu2+elution pre-staining paper chromatography-vis-spectrophotometry,NCE-PSPCVS)。该方法是将滤纸中的GABA斑点剪下,每个斑点用5mL 75%的乙醇溶液洗脱,待GABA完全洗脱后,用紫外分光光度计测定洗脱液在570nm处的光吸收值(A570)。此方法准确性高,线性关系良好,R2为0.994,相对标准偏差RSD为0.82%~1.72%,回收率96.95%~102.90%。经高效液相色谱法(HPLC)验证,此方法准确性高,可用于GABA定量测定。Gamma-aminobutyric acid plays an important role in the fields of food and pharmaceutics.Many methods have been employed to determine gamma-aminobutyric acid,including pre-staining paper chromatography-vis-spectrophotometry in which Cu^2+was described to stabilize ninhydrin-ninhydrin hydrate chromophore.In the current work,we report a new quantitative method to measure gamma-aminobutyric acid,termed as No-Cu^2+elution pre-staining paper chromatography-vis-spectrophotometry(NCE-PSPC-VS).The GABA spots were cut out from the paper and were eluted with 5mL 75%ethanol solutions.Afterwards the absorption values at 570 nm(A 570)of the eluents were read in a UV-vis spectrophotometer.This method is accurate with R^2=0.994,RSD ranging from 0.82%-1.72%,and recoveries between 96.95%-102.90%.The validation of this method was confirmed by HPLC,indicating that the developed approach could be an alternative of xisting methods for the determination of gamma-aminobutyric acid.
