摘  要:目的探讨人工寒潮促发糖尿病大鼠脑组织中细胞粘附分子的表达变化。方法采用糖尿病(diabetes mellitus,DM)模型大鼠24只和正常组大鼠14只,再分寒潮与非寒潮两亚组,通过免疫组化检测脑组织中ICAM-1、VCAM-1、P-选择素的表达水平,其余切片行HE染色。结果各组大鼠脑皮质中ICAM-1、VCAM-1、P-选择素等均有表达,P-选择素在丘脑和海马中也少量表达。正常对照寒潮组与正常对照非寒潮组比较,大鼠脑皮质中的ICAM-1、VCAM-1、P-选择素等阳性表达的光密度值无明显差异。DM组与正常对照组比较,DM组大鼠脑皮质中ICAM-1、VCAM-1、P-选择素等的光密度值均明显升高,差异具有统计学意义(P<0.05)。DM寒潮组与DM非寒潮病组比较,DM寒潮组大鼠脑皮质中ICAM-1、VCAM-1、P-选择素等的光密度值均明显升高,差异具有统计学意义(P<0.05)。结论寒潮是脑卒中发生的重要诱因,细胞粘附分子表达上调可能在寒潮促使脑卒中发生的发病机制中起着重要作用。Objective To study the change of artificial cold wave on the expression of cell adhesion molecules in the cerebral cortex in streptozotocin-induced diabetic rats.Methods 24 rats with type 2 diabetes mellitus and 14 normal rats were divided into cold wave exposure and non-exposed subgroups separately(n=8).then take one section from each eight to test ICAM-1,VCAM-1 and P-selectin in brain tissue by means of immunohistochemistry method,and make HE stain in the rest sections.Results 1 The neurons which have positive expressions of ICAM-1,VCAM-1,P-selectin in different groups of rats'cerebral cortex are significant,and the P-selectin also has obvious expressions in the hypothalamus and hippocampus.2 The expression of ICAM-1,VCAM-1 and P-selectin in the normal rats'cerebral cortex doesn’t ascend obviously in the exposure subgroup,and the result of statistical analysis is P>0.05 showing that there is no significant difference between exposure and non-exposed subgroups.3 The expression of ICAM-1,VCAM-1 and P-selectin in the diabetic rats'cerebral cortex in both the cold wave exposure and non-exposed subgroups ascend obviously higher then the normal groups,and the result of statistical analysis is P<0.05 showing that there are significant differences between them.4 The expression of ICAM-1,VCAM-1 and P-selectin in the diabetic rats'cerebral cortex in cold wave exposure subgroup ascend obviously higher then non-exposed subgroups,and the result of statistical analysis is P<0.05 showing that there are significant differences between them.Conclusion The up regulation of cell adhesion molecules may play an important role in the occurrence of stroke,and cold is an important external trigger.
