摘　　要：目的:观察环状RNAcirc-PDE3B(circ-PDE3B)在食道鳞癌中的表达及对人食道鳞癌细胞株ECA109增殖、迁移的影响。方法:采用高通量测序方法检测食道鳞癌组织和癌旁组织中的环状RNA的表达; MTT法检测siRNA干扰circ-PDE3B表达后对食道鳞癌细胞增殖的影响;平板克隆实验、划痕实验检测siRNA干扰circ-PDE3B表达后对食道鳞癌细胞成瘤、细胞迁移的影响。结果:在食道鳞癌细胞中circ-PDE3B表达升高,siRNA干扰circ-PDE3B的表达可抑制食道鳞癌细胞ECA109的增殖和迁移。结论:在食道鳞癌中,干扰circ-PDE3B的表达可抑制肿瘤细胞的增殖和迁移。Objective:To observe the expression of circular RNA circ-PDE3B(circ-PDE3B)in esophageal squamous cell carcinoma and its effect on proliferation and migration of human esophageal squamous cell carcinoma cell line ECA109.Methods:High-throughput sequencing was used to determine circular RNA expression in esophageal squamous cell carcinoma and tissues;MTT assay was used to determine the effect of siRNA on the proliferation of esophageal squamous carcinoma cells.Plate cloning experiments and wound healing assay were performed to detect the effect of siRNA on the expression of circ-PDE3B in the tumor formation and cell migration of esoph-ageal squamous cell carcinoma.Results:The expression of circ-PDE3B was elevated in esophageal squamous carcinoma cells,and siRNA interference with the expression of circ-PDE3B could inhibit the proliferation and migration of esophageal squamous cell carcinoma cell line ECA109.Conclusion:In esophageal squamous cell carcinoma,inhibition of circ-PDE3B expression inhibits tumor cell proliferation and migration.
