摘  要:以提高竹叶蛋白质得率为优化目标,采用响应面法对竹叶蛋白质提取工艺进行优化,考察提取次数、p H、温度、时间和液料比对竹叶蛋白质提取得率的影响。利用Box-Behnken试验和响应面分析法,研究了各变量及其交互作用对竹叶蛋白质得率的影响,模拟得到二次多项式回归方程的预测模型,得到最佳提取工艺。通过回归分析确定优化后的提取工艺参数为:提取2次,p H 6,温度33. 17℃,时间87.60 min,液料比23. 39 m L·g^(-1),蛋白质得率为7. 64%。此条件下,竹叶蛋白质提取得率的实测结果为7. 49%,与响应面拟合所得方程的预测值符合,比未优化的纯水提取竹叶蛋白得率提高了2. 621%。优化得出的回归模型具有一定的实际指导意义,为竹叶蛋白质提取的工厂放大和深入研究提供一定的实验研究依据。In order to improve the extraction yield of the protein from bamboo leaves, we optimized response surface methodology to investigate the effects of the extraction conditions including pH, temperature, duration and the extract-substrate ratio. According to the single factor experiments, the optimum conditions for extraction of bamboo leaf proteins was forecasted via Box-Benhnken central composite design and response surface methodology. The prediction model of quadratic polynomial regression equation was simulated. The results showed that the optimal conditions for extraction of bamboo leaf protein were as follows: extracted twice at pH 6, 33.17 ℃ for 87.60 min, and extract-substrate ratio of 23.39 mL·g^-1 . Under these conditions the yield of protein amounted to 7.64%. The experimental extraction yield of protein was 7.49%, with the relative error of 0.15% in comparison with the predicted value. This indicated that the optimized regression equation may derive practical significance, providing a theoretical and experimental reference for industrial extraction.
