摘 要:目的探讨天然牛磺酸对肝硬化门静脉高压(PHT)大鼠门静脉系统血流动力学及内皮细胞的影响。方法建立肝硬化PHT大鼠模型。将45只肝硬化PHT大鼠随机分为对照组、结合牛磺酸组(A组)及天然牛磺酸组(B组),每组15只。A组给予结合牛磺酸灌胃,B组给予天然牛磺酸灌胃,对照组给予等量0. 9%氯化钠溶液灌胃。21 d后,比较3组大鼠门静脉血流量(PVF)、门静脉压力(PVP)、门静脉阻力(PVR)、平均动脉压(MAP)以及肝脏血流灌注量(HPV)水平,并在超微电镜下观察门静脉内皮细胞超微结构。结果 A组及B组PVP、PVR、PVF均低于对照组(均P <0. 05);对照组、A组、B组MAP、HPV水平依次升高(P <0. 05)。电镜下可见对照组门静脉内皮细胞变性或脱落,基底膜完整性遭破坏,A组及B组内皮细胞结构完整,B组基底膜正常。结论天然牛磺酸可减轻肝硬化门脉高压大鼠门静脉压力,改善效应内皮细胞的超微结构,且其效果优于结合牛磺酸。Objective To investigate the effect of natural taurine on hemodynamic and endothelial cells in cirrhotic rats with portal hypertension(PHT).Methods The model of cirrhotic rats with PHT was established,and then 45 cirrhotic rats with PHT were randomly divided into control group,combined taurine group(group A),or natural taurine group(group B),with 15 rats in each group.Group A and group B were given combined taurine and natural taurine by gavage,respectively,while the control group received the same dose of 0.9%sodium chloride solution.After 21 days of intervention,the levels of portal venous blood flow(PVF),portal venous pressure(PVP),portal vein resistance(PVR),mean arterial pressure(MAP),and hepatic blood flow perfusion(HPV)were compared among three groups,and ultrastructure of portal vein endothelial cells was observed under ultra-micro electron microscope.Results The PVP,PVR and PVF were lower in group A and group B than in the control group(P<0.05),the levels of MAP and HPV increased successively in control group,group A and group B(P<0.05).Under the electron microscopy,degeneration or abscission of portal vein endothelial cells was observed,and the integrity of basement membrane was damaged in the control group,while the structure of endothelial cells was intact in the group A and group B,and basement membrane was normal in group B.Conclusion The natural taurine can significantly reduce portal pressure in cirrhotic rats with PHT,and improve the ultrastructure of endothelial cells,which is superior to combined taurine.
