摘 要:目的用免疫组化技术动态观察新西兰白兔脊髓空洞症模型中TGF-β1的表达情况,探讨其在脊髓空洞前状态形成中的作用机制。方法将90只健康、体重1. 5~2 kg的新西兰白兔(雌雄不限),随机分为Kaolin组、0. 9%氯化钠溶液组和对照组,每组30只。于术后第1、3、7、10、14天后处死,迅速完整取出C1-2节段4 mm,置4%多聚甲醛后固定48 h,供TGF-β1免疫组织化学染色检查。用HPIAS-1000高清晰度彩色病理图文分析系统对TGF-β1阳性反应物积分光密度值(IOD)进行半定量检测,进行统计分析。结果 TGF-β1免疫组织化学检测发现,对照组和0. 9%氯化钠溶液组动物颈髓灰质和白质神经元及胶质细胞均有弱阳性表达。Kaolin组动物于术后第1天上颈髓组织神经元表达有明显升高,神经胶质细胞亦可见阳性表达,主要为胞浆表达;第3天、7天、10天在上颈髓灰质及白质神经元、神经胶质细胞呈强阳性表达,胞浆可见粗的棕黄色颗分布,以第10天最为显著。第14天则TGF-β1表达稍微减弱。0. 9%氯化钠溶液组和对照组TGF-β1表达的IOD测定差异无统计学意义(P> 0. 05); Kaolin组动物颈髓组织中TGF-β1表达与0. 9%氯化钠溶液组和对照组相比明显升高,其中以第10天最为显著(P <0. 01)。结论本实验结果显示在脊髓空洞形成早期,脊髓组织TGF-β1表达上调,提示其在脊髓空洞前状态病理生理形成过程中可能发挥重要作用。Objective To observe the expression of TGF-β1 in presynrix state of rabbits and explore its action mechanism in the development of syringomyelia,especially in presyrinx state.Methods Ninety New Zealand white rabbits,with body weight:1.5~2kg,were randomly divided into Kaolin group(n=30),saline group(n=30)and control group(n=30).The rabbits were sacrificed on days 1,3,7,10 and 14 after surgery.A 4mm-long cervical cord was taken out from C12 segment and placed in 4%paraformaldehyde for 48h for immunohistochemistry to observe the expression of TGF-β1.HPIAS-1000 system was used for the semi-quantitative detection of IOD.Moreover,data obtained in this study were statistically analyzed.Results Immunohistochemistry assay showed weak expression of the gray matter around the central canal,white matter neuron and gliocyte of the rabbits were detected in saline group and control group.The expression of neurons in the upper cervical spinal cord at 1d after surgery in Kaolin group was significantly increased.And the positive expression(mainly nucleus expression)could be detected in glial cells.On days 3,7 and 10 after surgery,the strongly positive expression of the gray matter around the central canal,white matter neuron and gliocyte of the rabbits were detected,in which those at 10d were strongest.Coarse brown-yellow granules could be detected in cytoplasm.The expression of TGF-β1 at 14d became weaker.There were no significant difference in Integral optical density(IOD)of TGF-β1 between saline group and control group(P >0.05).Moreover,the expression levels of TGF-β1 of cervical spinal tissue in Kaolin group were significantly increased,as compared with those in saline group and control group,in which,those at 10d were most significant.Conclusion The up-regulation of the expression of TGF-β1 in spinal cord tissues indicated that TGF-β1 may play an important role in pathogenesis of presynrix state in rabbits.
