端粒酶逆转录酶mRNA的表达与非小细胞癌患者诊断和预后的相关性分析

摘　　要：目的检测人端粒酶逆转录酶(hTERT) mRNA在非小细胞肺癌(NSCLC)患者中的表达情况,评估其作为诊断和(或)预后因子的价值。方法 2011至2012年接受治疗的40例NSCLC患者,采用实时定量聚合酶链反应(QRT-PCR)检测12例女性和28例男性NSCLC患者癌性和非癌性肺组织中hTERT的表达情况,根据临床资料及1、2、5年生存率对结果进行分析。结果癌组织中的hTERT的表达显著高于无肿瘤浸润的肺实质(P <0. 05)。hTERT在肿瘤组织中的表达与年龄、性别、分级及临床分期无关;两种组织病理学类型(腺癌和鳞状细胞癌)间的hTERT表达有显著性差异(P=0. 01); hTERT在NSCLC组织中的表达与生存率无关。结论 QRT-PCR在肿瘤及相应无癌组织中检测到hTERT mRNA表达,可作为NSCLC的诊断指标,但它似乎不是NSCLC患者的预后因素。Objective To explore the expression of telomerase reverse transcriptase mRNA in non small cell carcinoma, and to evaluate its value in diagnosis and prognosis of patients with non small cell carcinoma (NSCLC). MethodsForty patients with NSCLC (28 males and 12 females), who were treated in our hospital from 2011 to 2012, were enrolled in the study. QRT PCR was used to measure the expression levels of hTERT in cancerous tissues and non-cancerous tissues. And the results were analyzed according to clinical data and the 1, 2, 5 year survival rates. ResultsThe expression levels of hTERT in cancerous tissue were significantly higher than those in lung parenchyma free from neoplasm infiltration ( P < 0.05 ). There was no correlation between the expression of hTERT in cancerous tissue and patient’s age, gender, grading and clinical stage,however, there were significant differences in the expression levels of hTERT between the two types of histopathological patterns (adenocarcinoma and squamous cell carcinoma)( P <0.05). In addition,there was no correlation between the expression of hTERT in NSCLC tissues and survival rate of patients. ConclusionThe expression of hTERT mRNA detected by QRTPCR in tumor and corresponding cancer-free tissues can be used as diagnostic indexes of NSCLC,but it seems not to be a prognostic factor.
