摘  要:目的探索miR-30d表达变化与赫赛汀治疗乳腺癌耐药性的关系。检测miR-30d是否调控HER2阳性乳腺癌细胞对赫赛汀的耐药性,并初步证实该效应是否与miR-30d调控细胞自噬过程相关。方法通过Q-PCR检测miRNA-30d在HER2阳性和HER2阴性乳腺癌组织以及乳腺癌细胞中的表达情况,向HER2高表达和HER2低表达的乳腺癌细胞中转染miR-30d mimics和miR-30d inhibitors,检测转染后细胞对赫赛汀的耐药性是否发生改变。通过Q-PCR检测miR-30d过表达和敲减后细胞自噬相关基因Beclin1和LC3的表达水平变化。结果HER2阳性乳腺癌组织和乳腺癌细胞中mir-30d表达显著下调(P<0.05);转染mir-30d能逆转HER2阳性乳腺癌细胞对赫赛汀的耐药性,同时下调细胞自噬相关基因Beclin1和LC3的表达水平。结论 MiR-30d的表达下调与HER2阳性乳腺癌对赫赛汀的耐药性相关。miR-30可能通过调控自噬相关基因的表达,改变乳腺癌细胞对赫赛汀的耐药性。Objective To explore the relationship between miR-30d expression and herceptin resistance in the treatment of breast cancer.To detect whether mir-30d regulates HER2 positive breast cancer cells'resistance to herceptin,and to preliminarily confirm whether this effect is related to mir-30d's regulation of cell autophagy.Methods The expression of miR-30d in HER2 positive and HER2 negative breast cancer tissues and breast cancer cells were detected by Q-pcr.Mir-30d mimics and mir-30d inhibitors were transfected into breast cancer cells with high HER2 expression or low HER2 expression to detect whether the cell resistance to herceptin changed after transfection.Changes in the expression levels of cell autophagy related genes Beclin1 and LC3 in mir-30d overexpression or knockdown cells were detected by Q-pcr.Results The miR-30d expression was down-regulated in HER2 positive breast cancer tissues and cell lines.Mir-30d transfection could reverse HER2 positive breast cancer cells'resistance to herceptin,and down-regulate the expression levels of Beclin1 and LC3 gene.Conclusion Downregulation of mir-30d expression was associated with HER2 positive breast cancer resistance to herceptin.Mir-30 may alter the resistance of breast cancer cells to herceptin by regulating the expression of autophagy-related genes.
