长链非编码RNA癌易感性候选基因2在卵巢癌中的表达及其对SKOV3细胞恶性表型的影响

摘　　要：目的探讨长链非编码RNA癌易感性候选基因2(LnCRNA CASC2)在卵巢癌中的表达及对卵巢癌SKOV3细胞恶性表型的影响。方法 q RT-PCR检测18例卵巢癌及正常癌旁组织;5株人卵巢癌细胞株A2780、Caov3、HO8910、SKOV3、OVCAR3和人正常卵巢上皮HOEpiC细胞LnCRNA CASC2的相对表达量;通过重组质粒在人卵巢癌SKOV3细胞中过表达LnCRNA CASC2,采用MTT法、流式细胞术、侵袭实验、Western blot检测过表达LnCRNA CASC2后SKOV3细胞增殖、凋亡、侵袭及Bcl-2、Bax蛋白表达情况。结果 LnCRNA CASC2在卵巢癌组织、卵巢癌细胞中表达水平低于对应的癌旁组织及卵巢上皮细胞(P <0.01)。LnCRNA CASC2重组质粒转染SKOV3细胞后可显著抑制细胞的增殖能力、侵袭能力,促进细胞凋亡(P <0.05)。过表达LnCRNA CASC2后,SKOV3细胞Bcl-2表达显著降低、Bax蛋白表达显著增加(P <0.05)。结论 LnCRNA CASC2在卵巢癌组织中表达降低,卵巢癌细胞中过表达LnCRNA CASC2后细胞恶性能力降低,其在卵巢癌中可能发挥抑癌基因的作用,值得进一步研究。Objective To investigate the expression of long non-coding RNA CASC2(LnCRNA CASC2)in ovarian cancer and its influence on malignant phenotype of ovarian cancer SKOV3 cells.Methods The relative expressions of LnCRNA CASC2 in 18 cases of ovarian cancer and normal paracancer tissues,in 5 human ovarian cancer cell lines A2780,Caov3,HO8910,SKOV3,OVCAR3 and human normal ovarian epithelium HOEpiC were detected by RT-PCR.LnCRNA CASC2 was over-expressed by recombinant plasmid in SKOV3 cells,then the proliferation,apoptosis,invasion and the expression of Bcl-2 and Bax protein were determined by MTT method,flow cytometry,invasive experiment and Western blot.Results The expression level of LnCRNA CASC2 in ovari an cancer tissues and ovarian cancer cells was lower than that in the corresponding paracancer tissues and ovarian epithelial cells(P<0.01).The proliferation and invasion of SKOV3 cells were significantly inhibited and the apop tosis was promoted by transfected recombinant plasmid LnCRNA CASC2(P<0.05).After the over-expression of LnCRNA CASC2,the expression of bcl-2 of SKOV3 cells decreased and Bax protein levels increased significantly(P<0.05).Conclusion The expression of LnCRNA CASC2 in ovarian cancer tissue is reduced.The malignant ability is reduced when the level of LnCRNA CASC2 increased,which may play the role of tumor suppressor genes and it deserves further research.
