摘 要:目的观察Ras鸟嘌呤核苷酸交换因子1(RasGRF1)在结直肠癌细胞的表达及对人肠癌细胞株HCT116增殖、凋亡和细胞周期的影响。方法采用RT-PCR方法检测结直肠癌细胞中RasGRF1的表达;MTT法检测RasGRF1对人结直肠癌细胞增殖的影响;Annexin V-FITC/PI染色流式细胞术检测对细胞凋亡和细胞周期的影响。结果 RasGRF1在人结肠癌细胞HCT116中过表达;siRNA干扰RasGRF1表达使人结肠癌细胞HCT116的增殖率降低;siRNA干扰RasGRF1表达使人结肠癌细胞HCT116的凋亡率增加并使细胞周期停留在G1期。结论在人结直肠癌细胞中RasGRF1是高表达的,siRNA干扰RasGRF1的表达可抑制人结肠癌细胞HCT116的增殖,凋亡率增加并使HCT116的细胞周期停留在G1期。Objective To observe the expression of RasGRF1 in colorectal cancer cells and its effect on proliferation,apoptosis and cell cycle in human colon cancer cell line HCT116.Methods Expression of RasGRF1 in colorectal cancer cells was detected by RT-PCR;RasGRF1 on the proliferation in human colorectal cancer cells was detected by MTT;PI staining on the cell cycle was detected by flow cytometry.Results RasGRF1 overexpressed in human colon cancer cell HCT116.Inhibition of RasGRF1 by siRNA reduced the proliferation rate in human colon cancer cell HCT116.The apoptosis rate of human colon cancer cell HCT116 was increased and left the cell cycle at G1 phase after inhibition of RasGRF1 by siRNA.Conclusions RasGRF1 is highly expressed in human colorectal cancer cells.siRNA interference RasGRF1 expression could inhibit the proliferation of human colon cancer cells HCT116,increase the rate of apoptosis and keep the cell cycle of HCT116 at G1 phase.
