摘 要:目的通过免疫共沉淀联合质谱分析筛查与单纯疱疹病毒1型(HSV-1)中US11蛋白存在潜在相互作用的宿主蛋白。方法使用荧光标记的HSV-1病毒感染人成纤维细胞(HFF),通过免疫共沉淀(IP)方法对US11及其相互作用蛋白进行提取和纯化,并与质谱(MS)联用来鉴定相关蛋白。结果利用IP-MS分析技术成功筛选出7个与US11存在潜在相互作用宿主蛋白,其中DNA甲基转移酶3A(DNMT3A)被定性为一种新的US11结合蛋白。结论 HSV-1病毒US11蛋白与哺乳动物DNA甲基转移酶机制的功能关联,DNMT3A是HSV-1感染所需的有效宿主因子。Objective To screen host proteins potentially interacting with US11 protein in herpes simplex virus type 1(HSV-1)by immunoprecipitation combined with mass spectrometry.Methods The HFF was infected with the fluorescently labeled HSV-1 virus,and US11 and its interacting protein was extracted and purified by co-immunoprecipitation,and mass spectrometry was performed to identify the related protein.Result IP-MS analysis technology was used to successfully screen 7 host protein with potential interaction with US11,DNA methyltransferases 3A was identified as a new US11 binding protein.Conclusion Our results highlight a functional association of virus US11 protein with the mammalian DNA methyltransferase machinery,indicating DNMT3A as a host factor required for effective HSV-1 infection.
