摘  要:为提高枯草芽孢杆菌菌株8-32抗真菌物质的产量,在单因素试验基础上,采用Plackett-Burman设计法找出影响菌株Bacillus subtilis 8-32产抗真菌物质的主要因素为培养时间、装液量和可溶性淀粉含量。采用最陡爬坡试验逼近最大响应区域,运用Box-Behnken试验设计及响应面分析法对3个显著影响因子的最佳配比进行优化,得到最佳发酵工艺参数为培养时间72.28h,装液量59.80mL/250mL三角瓶,可溶性淀粉含量1.51%。优化后抑菌圈直径从试验设计初期的14.21mm提高至24.51mm。In order to increase the yield of antifungal substances of Bacillus subtilis 8-32,Plackett-Burman method was used on the basis of single factor experiment,and the result showed that the main factors influencing the produce of antifungal substances were culture time,amount of liquid and soluble starch content.The steepest ascent experiment was used to approach the maximum response value,and the optimum ratio of 3 significant factors was optimized by Box-Behnken test design and response surface analysis.The optimum fermentation process parameters were 72.28 h culture time,59.80 mL/250 mL triangle bottle and 1.51%soluble starch.After optimization,the diameter of the inhibition zone increased from 14.21 mm at the beginning of the trial design to 24.51 mm.
