溴结构域蛋白4表达与肝癌患者预后的关系及其对HCCLM3细胞增殖和凋亡的影响

摘　　要：目的检测溴结构域蛋白4(BRD4)在肝癌组织中的表达,分析其表达与预后的关系,探讨沉默BRD4表达对肝癌HCCLM3细胞增殖和凋亡的影响。方法收集208例肝癌患者手术切除的肿瘤组织和癌旁组织,采用免疫组化法检测BRD4表达,比较BRD4高表达和低表达患者的肿瘤复发率、术后生存期。选取BRD4高表达的HCCLM3细胞,将其分为空白组、空载组、shRNA-1抑制组,shRNA-1抑制组转染对应的BRD4慢病毒抑制基因序列抑制BRD4表达,空载组加入慢病毒空载体及转染试剂,空白组只加入转染试剂。采用Western blotting法检测各组BRD4表达,CCK-8法检测细胞增殖情况,流式细胞术检测细胞凋亡率。结果肝癌组织中BRD4高表达率高于癌旁组织(P <0. 01)。BRD4高表达患者的肿瘤复发率较低表达患者升高,术后生存期较低表达患者缩短(P均<0. 01)。shRNA-1抑制组与其他两组相比较,细胞增殖OD值降低,细胞凋亡率升高(P均<0. 01)。结论高表达BRD4的肝癌患者预后较差;抑制BRD4表达可抑制肝癌细胞增殖,促进肝癌细胞凋亡。Objective To detect the expression of bromodomain protein 4(BRD4)in the hepatocellular carcinoma,analyze the relationship between its expression and prognosis,and to investigate the effect of silencing BRD4 on proliferation and apoptosis of hepatoma HCCLM3 cells.Methods The tumor tissues and adjacent tissues of 208 patients with liver cancer were collected.The expression of BRD4 was detected by immunohistochemistry.The tumor recurrence rate and survival time of patients with high and low expression of BRD4 were compared.The HCCLM3 cells with high expression of BRD4 were screened out and then were divided into the blank group,empty vector group and shRNA-1 inhibition group(the inhibition effect was obvious).The corresponding BRD4 lentivirus suppression gene sequence was transfected into the cells in the shRNA-1 inhibition group to inhibit its BRD4 expression,the empty vector group was added with lentivirus empty vector and transfection reagent,and the blank group was only added with transfection reagent.The expression of BRD4 in each group was detected by Western blotting.The cell proliferation was detected by CCK-8,and the apoptosis rate was detected by flow cytometry.Results The expression of BRD4 in the liver cancer tissues was higher than that in the adjacent tissues(P<0.01).Patients with high BRD4 expression had higher tumor recurrence rate and lower postoperative survival than patients with low BRD4 expression(both P<0.01).Compared with the other two groups,the OD value of the cell proliferation decreased and the apoptosis rate increased in the shRNA-1 inhibition group(both P<0.01).Conclusion The prognosis of liver cancer patients with high expression of BRD4 is poor,and the inhibition of BRD4 expression can inhibit the proliferation of hepatoma cells and promote the apoptosis of liver cancer cells.
