摘　　要：目的观察长链非编码RNA RP11-769O8.2在肝癌(HCC)组织、细胞系中的表达,评估其编码能力,预测其靶基因。方法采用实时荧光定量PCR法检测HCC组织、细胞系RP11-769O8.2,在线数据库评估RP11-769O8.2的编码能力及预测其靶基因。结果 HCC组织中RP11-769O8.2表达量低于癌旁组织,其表达水平与患者肝硬化程度有关(P均<0.05);HCC细胞系SK-Hep1中RP11-769O8.2相对表达量低于Huh7、Hep G2、97L(P均<0.05);RP11-769O8.2的编码能力评分为-1.28;RP11-769O8.2的靶基因为YES原癌基因1(YES1),HCC组织中YES1表达量高于癌旁组织,且与RP11-769O8.2的表达呈正相关(P均<0.05)。结论 RP11-769O8.2在HCC组织中低表达,与患者的肝硬化程度相关,编码能力弱;YES1在HCC组织中低表达,与RP11-769O8.2表达呈正相关。Objective To observe the expression of long non-coding RNA(lncRNA)RP11-769O8.2 in the hepatocellular carcinoma(HCC)tissues and cell line,and to evaluate its coding ability and predict its target gene.Methods The real-time polymerase chain reaction(qRT-PCT)was used to detect the RP11-769O8.2 in the HCC tissues and cell line,and online database was used to evaluate the coding ability of RP11-769O8.2 and to predict its target gene.Results The relative expression of RP11-769O8.2 RP11-769O8.2 in the HCC tissues was significantly lower than that of the adjacent tissues,and the decrease of its expression level was related to the degree of cirrhosis of patients(P<0.05).The relative expression of RP11-769O8.2 in the HCC cell line SK-Hep1 was significantly lower than that of Huh7,Hep G2,and 97 L(all P<0.05).RP11-769O8.2 RP11-769O8.2’s coding ability score was-1.28;the online database predicted that the target gene for RP11-769O8.2 RP11-769O8.2 was Yes proto-oncogene 1(YES1);the expression of YES1 in the HCC tissues was higher than that of the adjacent tissues,and was positively correlated with the RP11-769O8.2 expression(all P<0.05).Conclusion RP11-769O8.2 is lowly expressed in the HCC tissues,which is related to the degree of cirrhosis of patients,and its the coding ability is weak;YES1 is lowly expressed in the HCC tissues,which is positively correlated with the expression of RP11-769O8.2.
